Objective: Obtaining new born tissues from distraction areas and rapid growth bone tissues from new born dogs by establishing the model of distraction osteogenesis of dogs' mandibles. And conducting the study of lncRNA expressions on these objects using high-throughput sequencing technologies.Then compare the similarities and differences of lncRNA biology information expressions in distraction osteogenesis and in rapid growth osteogenesis.Method: In this experiment, the objects are divided into three groups.Group DO is experimental group, which is the distraction of adult dogs'mandibles. Giving a single line distraction surgery on adult dogs' mandibles and collecting the new born tissues from distraction areas immediately after finishing the operation. Group P is control group with the participant of new born dogs. Killing the dogs immediately after birth and after two weeks of their birth, and then collecting the jaw bone tissues. Group N is blank group with mandible tissues of healthy adult dogs. Collecting RNA from the three different kinds of tissues and removing rRNA, and then comparing the similarities and differences among the three through high-throughput sequencing.Results: HE staining: the bone tissues of distraction osteogenesis and embryo osteogenesis are both cartilage tissues with abundant blood vessels, cells and collagen fibers. The cellular composition and tissue composition are similar in these two groups. Biological analysis of lncRNA: there are 225 lncRNA and 67 target genes relating to the formation and growth of vessels. 160 lncRNA and 38 target genes are angiogenesis function-related. Moreover, 179 lncRNA and 50 target genes are vasculogenesis function-related. Among all the lncRNA expressions related with vessels, the expressions of distraction period and two weeks' new born dogs are highly similar; compared with group N, the trend of lncRNA expressions in group DO-2 differs most with other groups.Conclusion: lncRNA may positive regulates IL-6 and promotes angiogenesis by upregulation. lncRNA may positive regulates SFRP2 and inhibits angiogenesis of distraction osteogenesis by upregulation.
