Objective : The insulinoma originated from neuroendocrine cells and peptidergic neurons, also known as functional pancreatic neuroendocrine tumors of pancreatic ? cells. The tumor is characterized by the abnormal secretion of insulin, which is caused by the abnormal secretion of insulin, which is not affected by the blood glucose level.Betatrophin is ANGPTLs (Angiopoietin-like proteins) after ANGPTL1?7 of the eighth new members, mainly expressed in human and mouse the liver and adipose tissue, this can be detected in the blood circulation of a secreted protein, because the structure and angiogenesis - like protein family members ANGPTL3 and ANGPTL4 have the same N terminal domain up to 20% also known as angiogenesis hormone like protein eight.Betatrophin protein was first discovered in human insulin the model can promote the resistance specificity of polypeptides proliferation and insulin secretion from pancreatic ? cells. Inspired by this, we first tested for betatrophin and insulinoma,looking for a potential breakthrough for the pathogenesis of cancer research. This experiment firstly according to the latest version of the World Health Organization digestive system tumor classification standards of tumor samples for pathologic diagnosis and classification of strict,Subsequently,We used immunohistochemistry to detect the betatrophin in insulinomas' tissues and normal controls, and to compare the groups by semi quantitative analysis. Finally, we also performed double labeling immunofluorescence detection of betatrophin in tumor tissues. To understand and explore the relationship between betatrophin and insulinoma.Methods: Samples were selected from 2009.1 to 2014.10 of the First Affiliated Hospital of Guangxi Medical University of the patients who were treated with surgical resection of the pancreatic specimens. The samples were grouped as follows: human insulinomas, group, normal human pancreas group,normal human liver group;WHO digestive system tumor classification criteria(2010) as a reference,improve the pathological diagnosis and pathological grading of insulinomas,Secondly,using immunohistochemical detection of betatrophin on insulinomas and control group,and then to convert the results of immunohistochemistry into MOD value (mean optical density) by using image analysis software, quantitative comparison after grouping, Ultimately, we carried out location detection of insulin and betatrophin by double labeling immunofluorescence method.Results: Pathological diagnosis of tumor: 25 cases of insulinoma (including 16 cases of stage G1, 9 cases of G2), and 2 cases of stage G3). Betatrophin in normal pancreatic tissue almost no expression, insulinomas' group expression compared with normal pancreatic tissue were significantly increased, P < 0.01;Betatrophin expression in insulinoma was not correlated with pathologic grade,benign and malignant diagnosis, age, gender, and disease duration (P>0.05).Double labeling immunofluorescence results suggest that betatrophin expression was significantly higher in tumor tissue of insulin, all the positive region of insulin in the cytoplasm were significantly, broad, more uniform expression betatrophin, a little insulin tumor tissue powder in non insulin positive cytosolic region also express different levels betatrophin.Conclusions: Betatrophin is abnormally high expression in insulinomas,and its mainly expressed in the cytoplasm of ? cells, which suggests that high concentration of' betatrophin may be involved in the occurrence and development of insulin tumor.
