Objective To investigate the expression of histone deacetylase 2(HDAC2)in patient with lung adenocarcinoma,and discuss relationship between the expression of HDAC2,interleukin-8(IL-8)and tumor necrosis factor-?(TNF-?)and tobacco smoke exposure and airway inflammation.Materials and Methods The 73 cases with lung adenocarcinoma and 14 cases of normal lung tissues(The 14 cases of normal lung tissues were collected from surgical patients who were not suffered lung cancer and without tobacco smoke exposure and COPD,including inflammatory pseudotumor,hamartoma,tuberculoma and so on)confirmed by pathological examination after surgical removals were collected in the First Affiliated Hospital and Affiliated Tumor Hospital of Guangxi Medical University from April 2014 to March 2015.All patients were taken preoperative peripheral venous blood 5 ml and collected lung adenocarcinoma tissues,para-cancer tissues and normal lung tissues after surgery.All patients were detected preoperatively with lung function test.People who have smoked more than 100 cigarettes or breathed the fumes of cigarettes at least one day a week(more than 15 minutes every day)more than three years are defined as tobacco smoke exposure.According to the lung function and whether tobacco smoke exposure or not,the 87 cases of patients were divided into four groups: group A: normal control group(14 cases);group B: without tobacco smoke exposure and chronic obstructive pulmonary disease(COPD)group(19 cases);group C: tobacco smoke exposure without COPD group(33 cases);group D: tobacco smoke exposure with COPD group(21 cases).The levels of HDAC2,IL-8 and TNF-? mRNA in lung adenocarcinoma,para-cancer tissues and peripheral blood mononuclear cells(PBMCs)were detected by real-time polymerase chain reaction(PCR)and the expression of HDAC2 protein was detected by western blotting,ELISA was used to detect the content of IL-8 and TNF-? in serum.Results(1)Compared with lung tissues of group A,the expression of HDAC2 at mRNA and protein levels in para-cancer tissues in group B has no statistics significance(P>0.05),and in group C and group D the expression of HDAC2 at mRNA and protein levels in para-cancer tissues were significantly lower(all P<0.05),and in group D decreased most obviously.Compared with the corresponding para-cancer tissues of each group,the expression of HDAC2 at mRNA and protein levels in lung adenocarcinoma tissues were significantly lower(all P<0.05).Compared with the lung adenocarcinoma tissues of group B,the expression of HDAC2 at mRNA and protein levels in lung adenocarcinoma tissues of group C and D were significantly lower(all P<0.05),and in group D decreased most obviously.Compared with lung tissues of group A,the content of IL-8 and TNF-? at mRNA levels in lung adenocarcinoma tissues and paracancer tissues of each group was significantly higher(all P<0.05),and the upregulation was more obvious in group B and most obvious in group D.Compared with the corresponding para-cancer tissues of each group,the expression of IL-8 and TNF-? at mRNA levels in lung adenocarcinoma tissues were significantly higher(all P<0.05).Compared with the lung adenocarcinoma tissues of group A,the expression of IL-8 and TNF-? at mRNA levels in lung adenocarcinoma tissues of group C and D were significantly higher(all P<0.05),and the up-regulation was more obvious in group D.(2)Compared with peripheral blood mononuclear cells(PBMCs)of group A,the expression of HDAC2 at mRNA levels in PBMCs of group B has no statistics significance(P>0.05)and in group C and D were significantly lower(all P<0.05),which in group D decreased most obviously;Compared with PBMCs of group B,HDAC2 at mRNA levels in group C and D were significantly lower(all P<0.05).Compared with PBMCs of group A,the expression of IL-8 and TNF-? at mRNA levels in PBMCs and the content of IL-8 and TNF-? in serum of each lung adenocarcinoma group was significantly higher(all P<0.05),and the up-regulation was more obvious in group C and most obvious in D.Compared with PBMCs of group B,the expression of IL-8 and TNF-? at mRNA levels in PBMCs and the content of IL-8 and TNF-? in serum of group C and D were significantly higher(all P<0.05).(3)The relative expression of HDAC2 in lung adenocarcinoma tissues and PBMCs showed no significant differences with respect to age,gender and TNM stage(P>0.05).IL-8 and TNF-? in lung adenocarcinoma tissues,PBMCs and serum showed no significant differences with respect to age and gender(all P>0.05),and in different classes of TNM stage,stage ? is higher than stage ?and?(all P<0.05),stage ?compared with stage? show no significantly difference(P>0.05).Conclusions(1)The expression of HDAC2 is down-regulated and IL-8,TNF-? are up-regulated in lung adenocarcinoma tissues,which may be closely related to the occurrence and development of lung adenocarcinoma.(2)In lung tissues and PBMCs of lung adenocarcinoma,tobacco smoke exposure causes the expression of IL-8 and TNF-? up-regulated,combined with COPD is more obvious,which mechanism may be associated with lower HDAC2 expression;(3)Airway inflammatory response in lung adenocarcinoma is increased and at the late stage is stronger than the early stage.
