PurposeExpression of HJURP(Holliday Junction Recognition Protein)protein in HCC specimen and peri-cancer tissues respectively,were detected with reversetranscription-polymerase chain reaction (RT-PCR) . To investigate the significance of expression of HJURP mRNA in HCC, and analyze its relationship with the clinicopathological parameters and the time of recurrence and metastasis after operation, to provide a scientific basis for the comprehensive treatment of hepatocellular carcinoma postoperative recurrence.MethodDuring March 2007 to December 2013 in Department of hepatobiliary surgery, the Affiliated Tumor Hospital of Guangxi Medical University underwent radical resection of 73 cases of postoperative patients with HCC tumor tissue, and take > 1cm cancer adjacent tissues of 21 cases as control(Among them, 19 cases had corresponding cancer tissues and adjacent tissues),no any anticancer therapy before surgery, all patients to exclude other tumors and pathological diagnosis with non hepatic cell carcinoma. The general clinical data of 73 cases were collected, including sex, age, family history, serum marker,tumor size, tumor size, tumor size, vascular tumor thrombus, pathological grading, and so on, with follow up.HJURP mRNA expression in liver cancer tissues and adjacent tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR) , HJURP protein in hepatocellular carcinoma and adjacent tissues was detected by Western blot. SPSS 16.0 statistical software was applied for data analysis. The correlation between the expression of HJURP mRNA and the clinical pathological characteristics of HCC were analyzed by Chi-square test.The time to recurrence or metastasis of HCC patients treated with radical resection were computed by the Kaplan-Meier method and compared by the Log-tank test .The conventional prognostic factors were analyzed by Cox regression model. A p-value less than 0.05 was condidered significantly.Results1?73patients were followed up successfully to March 11, 2016,,the longest follow-up time was 94 months, the shortest six month.35 cases with recurrence,metastasis and recurrence metastasis rate was 47.9%, the median time of recurrence was 9 months, both for intrahepatic recurrence.2?According to HJURP mRNA in liver tissue of the relative expression of the median value, 73 cases of liver cancer cases were divided into high expression group and low expression group. The relative expression of HJURP mRNA in 73 cases of HCC was 1.17, the median of HJURP mRNA relative expression in 21 cases of peri-cancer tissue was 0.15,the difference was statistically significant (P<0.05).In 73 cases of HCC, the high expression rate of HJURP mRNA in HCC was 50.7% (37/73). And in 21 cases of peri-cancer tissue, There were 1 cases of HJURP mRNA high expression (1/21). The high expression rate of HJURP mRNA in HCC organization was significant higher than that of peri-cancer tissue, The high expression rate of mRNA HJURP in 19 cases of HCC was significantly higher than that in the corresponding adjacent tissues(P<0.05).3.The expression rate of HJURP mRNA in difference gender,age,first-degree family history of HCC, hepatitis B virus statue, liver cirrhosis,number of tumor , vascular invasion , BCLC staging , TNM stage and so on,respectively,was not significantly different(P>0.05). Meanwhile, the relative expression of HJURP mRNA was related to preoperative serum ALT,preoperative AST,preoperative AFP levels, tumor size, vascular tumor thrombus,and the difference was statistically significant (P<0.05).4.To postoperative 6M,12M,18M and 24M for the group boundaries.Recurrence and metastasis time<6Mgroup, 6.1M?12M group?12.1M-18M group?18.1M?24M group and >24M group ,HJURP mRNA expression rate was35.7%?66.7%?75.0%?25.0%?50.0%, respectively,and the difference was no statistically significant (P > 0.05) . Recurrence and metastasis time <12M group and >12M group, ?18M group and >18M group, ?24M group and >24M group,?6m group and >12M group, ?6M group and >18M group, ?6m group and >24M group, ?12m group and >18M group,?12m group and >24M group,?12M group, 12.1M?24M and >24M group, ?6M group and >6M group,12.1M?24M group and >24M group, the HJURP mRNA high expression rate was no statistically significant (P > 0.05).5?Univariate analysis showed that tumor>5cm (P < 0.05 ) is a risk factor for recurrence of liver cancer after radical resection.. tumor>5cm group median recurrence time was 11.0 months, significantly shorter than the corresponding control group 31.5 months (P < 0.05); In TNM stage, the median recurrence time of phase?, phase ? and phase ? was 20.0 months, 17.0 months and 6.0 months, respectively, and the difference was statistically significant (P<0.05);HJURP mRNA high express group median recurrence time is 15.0 M, while HJURP mRNA low expression group was 22.0 M, but the differences have no statistical significance (P>0.05). Time of metastasis and recurrence was no significant statistical difference in this group such as age,gender, family history,drinking, liver cirrhosis, preoperative AFP, preoperative ALT, preoperative AST,tumor number, vascular invasion, vascular tumor thrombus, BCLC stage and so on(P>0.05)6?Cox regression model of Multivariate analysis of prognostic factors in HCC showed that tumor size is independent risk factors influence the post-operation relapse and metastasis of HCC patients.Conclusion1?HJURP mRNA protein expression had significant differences in HCC organization and peri-cancer tissue, expression level of HJURP mRNA in HCC was obviously higher than in peri-cancer tissue.2?The expression of HJURP mRNA in HCC was not significantly correlated with gender, age, family medical history, type B viral hepatitis,preoperative serum of AFP, number of tumor mass, vascular invasion,BCLC staging respectively(P>0.05, respectively). The expression of HJURP mRNA in HCC was significantly correlated with preoperative serum of ALT? AST, size of tumor mass , vascular tumor thrombus and TNM stage (P<0.05).3? The expression of HJURP mRNA was not related to the recurrence and metastasis of hepatocellular carcinoma after operation.4? The size of tumor was independent risk prognostic factors of HCC.
