Objective : To investigate the effects of finasteride on proliferation inhibition and migration of human prostate cancer PC-3 cell.Methods:According to the requirements of the program were to set up the experimental group and the control group,use finasteride effect as experimental group,without non finasteride effect as control group.The inhibition of cell proliferation was detected by CCK-8 kit.The migration of cell was detected by cell wound scratch assay.The expression of VEGF and HIF-1a were detected by ELISA kit.The expression of androgen receptor(AR)was detected by flow cytometry.Results: Cell viability was not significantly affected by finasteride exposure.Cell migration was inhibited by 60umol/ml and 100umol/ml of finasteride.The secretion of VEGF and HIF-1a was significantly lower than that of the control group.The expression of androgen receptor in human prostate cancer PC 3 cells was not affected by finasteride and the expression of androgen receptor in PC-3 cells could not be induced.Conclusions: Although our study find that finasteride can not inhibit the viability of PC-3 cell,but our findings indicate that finasteride may attenuate tumor aggressiveness and invasion,it may be through down-regulation of PC-3 secretion of vascular endothelial growth factor,hypoxia inducible factor-1a.At the same time,we also found that prostate cancer PC-3 cells did not express androgen receptor,finasteride not induced the expression of androgen receptor.
