Objective:To explore analgesic action of the exogenous neuropeptide substance galanin receptor agonist M1145 on rat lateral habenular nucleus.Research Methods:Based on behavioural algesthesia testing methods,to perform microinjection of agonist and antagonist of different concentration of galanin receptor? agonist(M1145),ntagonists(M871)and PKC pathway blockers(Chelerythrine)measure and record the rat caused by thermal stimulation and mechanical stimulation of HWL,which will be used as measurement indicators of pain,so as to explore if there exists analgesic action of galanin and its receptor on rat lateral habenular nucleus.Results: The lateral habenula nucleus in normal rats injected with different concentration GalR ? agonist M1145,the HWL by mechanical stimulation and thermal stimulation,and the best effect in 15 th minute.While injecting GalR ?antagonist M871 can partially blocking Gal and M1145 analgesic action.Injection of PKC pathway blockers Chelerythrine can inhibit the analgesic effect of Gal,prompt galanin analgesia mechanism by Gi/o G protein.Injection M1145 increased HWL in the rat model with chronic pain,and Compare the analgesic action anormal rats and contrast rats with chronic pain,there were no significant difference.In normal rats and neuralgia lateral habenula nucleus of rats injected with different concentration GalR ?agonist M617,nociceptive information transmission can be suppressed,and the analgesic effect without obvious difference;Compared with the effect of analgesia M1145 also do not have obvious difference,prompt Gal R ? and GalR ? are involved in the analgesic effect of neuralgia in rats.
