Objective:The study attempt to detect the expression of ELMO1 in human gastric cancer tissue,and analyze the association and meaning of clinical pathological features of gastric cancer,for the different expression of ELMO1.In vitro,to explore whether the different expression of ELMO1 affects ability of invasion and metastasis of gastric cancer,or not.Methods:1.Immunohistochemical(IHC)test is applied to detect the positve expression rate and location of ELMO1 in the gastric cancer tissues.Analyze the association and meaning between the expressional level of ELMO1 and clinical pathological features of gastric cancer.The real-time quantitative PCR and Western Blot were employed to detect expressional level of ELMO1 in gastric tissue and cells.And select the gastric cancer cell lines,which express highest level of ELMO1.2.After ELMO1 gene was silenced by ELMO1-siRNA,Wound-healing,transwell and MTS assay were used to test ability of migration and invasion of SGC7901,in vitro.Results:1.Immunohistochemical analysis manifest that the positive expression rate of ELMO1 is significantly higher in gastric cancer tissues than normal gastric tissues.And the more development of tumor pathologic stage,the more expression of ELMO1.What’s more,ELMO1 locates on cell membrane and cytoplasm of the gastric cancer.Moreover,the expressional level of ELMO1 is meaningfully related to tumor size,depth of invasion,pathologic differentiation degree,lymph node metastasis,distant metastasis and TNM stage,but not to gender or age.2.Western blot and qRT-PCR analysis display that expressional level of ELMO1 is significantly higher in gastric cancer tissues and cells than in normal gastric tissues and cells.3.Wound-healing,transwell and MTS assays show that inhibit the expression of ELMO1 can significantly reduce ability of the invasion and metastasis in gastric cancer cell.Conclusion:1.ELMO1 expresses highly in gastric cancer.The expressional level of ELMO1 is meaningfully related to tumor size,depth of invasion,pathologic differentiation degree,lymph node metastasis,distant metastasis and TNM stage,but not to gender or age.2.The expression of ELMO1 could increase ability of the invasion and metastasis in gastric cancer cells.
