Objective: To observe the effects of recombinant human erythropoietin(rhEPO)on the expression of JAK2 and STAT5 in lung tissue of the development of bronchopulmonary dysplasia(BPD)model,aimed to explore the protective mechanism of rhEPO on lung tissue of BPD model could be associated with the activation of jak2/stat5 signal pathway.Methods: Lipopolysaccharide(LPS)was used to cause the intrauterine infection of SD newborn rats,and then placed them at 60 percent high-oxygen environment for manufacturing the BPD model.The spra-gue-dawley(SD)rats were injected LPS or NS to gestational sacs on 15 th day of gestation,18 newborn pups produced by the normal pregnant mice treated with NS were randomly selected as air control group(group A).The remaining newborn mice produced by the pregnant mice treated with LPS were placed at the high oxygen environment for manufacturing the BPD model,and were randomly selected 36 pups divided into two groups,each groups had 18 newborn pups,namely the control group(group B),rhEPO group(group c),and group c were treated with rhEPO intervention,The drugs were given subcutaneously in the back of the BPD model.Before rhEPO intervention,6 newborn pups from NS and LPS produced pregnant rats were randomly selected,after anesthetizing them,we choose the lung tissue and stained them by HE,and observed the pathological changes of lung tissue.Next,the first administration time was recorded as the 1st day,7th and14 days later,Random number table were used to select 6 pups from each group,after anesthetizing them,we choosed their lung tissue and placed at liquid nitrogen,and transferred to-80℃ ultra-low temperature refrigerator;RT-PCR and Western blot assay were used to detect the expression levels of jak2,stat5 mRNA and protein in lung tissue of newborn rats BPD model treated with recombinant human erythropoietin intervention.Results:(1)The results showed the expansion,congestion of pulmonary capillary,and infiltration of inflammatory cells in the alveolar septa and alveolar in the lung tissue of newborn rats produced by group B,when compared to group A.(2)The jak2 and stat5 mRNA and protein expression levels were significantly lower in group B than normal at the group A,while the expression level of jak2 and stat5 was significantly increased in group C;(3)The differences between each group were statistically significant on 1st,7th and 14 th day of hyperoxia exposure(P<0.05)。Conclusions:(1)The BPD model of SD mice was successfully modeled.(2)The protective effect of rhEPO on lung tissue of BPD model is related to the regulation of Jak2/Stat5 signaling pathway and up regulation of its expression.(3)With the time of administration,the expression of Jak2 and Stat5 increased gradually.It was speculated that exogenous rh EPO promoted the activation of Jak2/Stat5 signaling pathway in a time-dependent manner.
