Objective:Through to the planting of lung cancer A549 cells into nude mice before radiotherapy infusion of hyperoxic liquid improve tumor hypoxia environment,enhance tumorsensitivity to radiotherapy,and proved to be in a high-oxygen environment reduced hypoxia-inducible factor-1??HIF-1??expression,slow tumor growth.Method:A549 cells were seeded in 30 only 46 weeks male nude mice,subcutaneous tumor volume to be up at around 80-150 mm3 by tumor volume and tumor-bearing mice weighingbalance principle divided into three groups,each 20Gy radiotherapy.Group A before radiotherapy about 30min using 20mg/kg to high oxygen liquid intravenous injection;B groupwith normal saline instead of high oxygen liquid;C negative group without any drug infusion before radiotherapy.Radiation immediately after the completion of each mouse arterial blood 0.2mL,check the arterial blood gas analysis and to ELISA and RT-PCR for detection of HIF-?alpha and VEGF expression.Growth delay time to observe the effects ofA,B and C group,tumor volume and tumor to radiotherapy,1 weeks after the end of radiotherapy the nude mice were killed and isolated from tumor tissue,the weighing and measuring diameter,and the data were statistically analyzed.Result:Arterial blood gas analysis showed that hyperoxia liquid group?A?and oxygen partial pressure?PO2?were significantly higher than those in the saline group?A?andnegative group?C?,pH value of hyperoxia liquid group?A?and physiological saline group?B?had no significant difference,but higher than that of negative group?C?;ELISA detection the expression level of hyperoxia liquid group of HIF-1 alpha and VEGF were significantly higher than that of saline group,negative group;RT-PCR results suggestedthat hyperoxia liquid group HIF-1alpha and its downstream target gene transcription level of VEGF was significantly lower than that of saline group,negative group.Conclusion:The high oxygen liquid infusion after planting nude mice significantly increased PO2 and PH value,oxygen-rich blood through the blood circulation into the tumor tissue,improving the microenvironment of tumor hypoxia induced HIF-1?and decrease of hypoxia the number of tumor cells and enhance the sensitivity of tumor cells to radiation;HIF-1?down while inhibiting the expression of their target genes VEGF,thereby inhibiting angiogenesis,slowing tumor growth and metastasis.
