To investigate the expression of X-linked inhibitor of apoptosis protein(XIAP),nuclear factor kappa B(NF-?B)protein and mRNA in osteosarcoma(OS),giant cell tumor of bone(GCTB)and osteochondroma(OC).To explore the relationship between the expression of XIAP,NF-?B protein and mRNA and the clinicopathological parameters of OS.To analyse the correlation between XIAP and NF-?B in OS,and provide the theory evidence for the research on occurrence,development,invasion and metastasis mechanism of OS.All pathological samples of 36 cases with OS,20 cases with GCTB and 20 cases with OC were collected from the Department of Pathology of the 251 st Hospital of PLA from January 2005 to January 2016.The diagnosis of 76 cases with bone neoplasms should be based on a combination of clinical evaluation,imaging characteristics and histopathological feature.The tissue did not receive radiation and chemotherapy before specimen collection.The expression of XIAP,NF-?B protein and mRNA were detected in OS,GCTB and OC by immunohistochemistry(IHC)and in situ hybridization(ISH)method.The relationships between the expression of two markers and the gender,age,tumor position,tumor size and pathological type,differentiation degree,Enneking stage,soft-tissue invasion and distant metastasis of OS were analysed.The correlation between XIAP and NF-?B in OS was analysed by Spearman rank correlation.The consistency of the expression of XIAP,NF-?B protein and mRNA in OS was analysed by Kappa test as well.There was positive expression in different degrees of XIAP protein and mRNA in OS and GCTB and no expression in OC.There was positive expression in varying degrees of NF-?B protein and m RNA in OS,GCTB and OC.The positive expression rates of XIAP protein and m RNA in OS,GCTB and OC were(69.44%,66.67%),(35.00%,25.00%)and(0,0)respectively,the expression of XIAP among bone neoplasms tissue had significant difference(P all<0.05).The positive expression rates of NF-?B and mRNA were(72.22%,69.44%),(45.00%,40.00%)and(15.00%,5.00%)respectively,the expression of NF-?B had significant difference as well(P all<0.05).The expression of XIAP,NF-?B protein and mRNA in OS was not correlated with the gender,age,tumor position,tumor size and pathological type(P all>0.05),but correlated with differentiation degree,Enneking stage,soft-tissue invasion and distant metastasis(P all<0.05).The expression of XIAP protein was positively correlated with NF-?B protein in OS(P<0.05).The expression of XIAP mRNA was positively correlated with NF-?B mRNA in OS as well(P<0.01).The expression of XIAP protein was consistent with XIAP mRNA in OS(P<0.01).The expression of NF-?B protein was consistent with NF-?B m RNA in OS as well(P<0.01).XIAP and NF-?B were highly expressed in OS and there was no difference between protein and mRNA.There was positive correlation between XIAP and NF-?B.These suggesting that XIAP and NF-?B may participate in the the occurrence and development of OS together.XIAP and NF-?B may have a synergistic interaction and promote the invasion and metastasis of OS.The combined expession of XIAP and NF-?B could be better to determinie the malignant level of OS.
