Objective : Research the biological effects of human MEC-1 Cells simulated by Microgravity and X-ray Radiation and exploration of the feasibility for clinical practice.Methods: Group: control group(c),X-ray irradiation group(X),Simulated microgravity group(M),X-ray irradiation+Simulated microgravity group(X+M).The irradiation dose was 0,0.5,1,2,4Gy.Based on CCK-8 to observeX+M group and X group MEC-1 cells,related index and to analyze the changes of its proliferateability;The flat cell clone formation experiment calculated their clone formation rate to reflect the difference growth activity of MEC-1 cell;Using the Transwell cell invasion experiment analyze the invasive ability ofMEC-1 cell;Counting the proportion of cell cycle by Flow cytometry instrument explore the meaning.Results:1)The OD value of MEC-1 cells in X+M group were 0.559,0.489,0.474,0.357,0.321,which were lower than X group at 24h(P<0.05).2)The clone formation rate of MEC-1 cells in X+M group were 383%,302.33%,166.67%,85.33%,41.34%,which were lower than X group(P <0.05).3)The number of invasive cells in X+M group were 102.67,90,72,55,39.33,which were less than X group at 24h(P<0.05).4)The G2/M phase of MEC-1 cells in the X+M group was significantly lower than X group(P<0.05).The proportion of G2/M were 25.8%,30.03%,34.8%,50.96%,59.57%,significantlycompared with those in X group(P<0.05).Conclusion:Simulated microgravity effect and X-ray radiation on MEC-1 cell can play an inhibitory role in proliferation and invasive abilityand enhanceits radiosensitivity.It also can provideexperimental basis for clinical practice.
