Association Of ABCB1 Gene Polymorphisms With Aspirin Resistance In Patient With Ischemic Stroke

Objective To investigate the single nucleotide polymorphisms of Multi-drug resistance(ABCB1/MDR1)gene with Aspirin resistance(AR)in patients with ischemic stroke.Methods This study was conducted 300 patients who were hospitalized in the Department of Neurology of the Affiliated Hospital of Qingdao University during the period from September 2014 to May 2016.All patients in the group met the diagnostic criteria of Chinese acute ischemic stroke diagnosis guidelines and were clinically diagnosed with an MRI or CT scan,and All patients were regularly treated by oral aspirin(100mg)for more than 7 days.According to the result of blood clots elastic figure(TEG)will be divided into the group of aspirin resistance(AR)75 cases(AA<50%)and aspirin sensitive(AS)225cases(?50%).Detected the genotype by restriction fragment length polymorphism(PCR–RFLP)and direct sequencing methods.Discussing the relationship between polymorphisms with haplotype and AR in patients with ischemic stroke.Results In this study the T allele frequency of C3435 T in AR group and AS group was57.3% and 43.1% respectively,the difference was statistically significant(?2=9.143,p=0.002).and the number of genotype CC+TT in AR group was more than AS group,the difference was also significantly(?2=4.369,p=0.037);There was no difference between C1236 T and G2677T/A in allele frequency and genotype distribution in the two group;Analyzed the haplotype was constituted by the three loci,the OR value of haplotype(C-T-T)was 1.602(?2=5.374,p=0.02),the OR value of haplotype(C-G-C)was 0.495(?2=8.775,p=0.003),the OR value of haplotype(T-G-C)was3.010(?2=5.846,p=0.015),while the OR value of haplotype(T-T-T)was 3.308(?2=4.650,p=0.031),the difference is statistically significant,and the risk of AR in patients with ischemic stroke can be increased by haplotype(C-T-T,T-G-C,T-T-T).Conclusion The polymorphisms of C3435 T in the ABCB1 gene have a relationship with AR in the patients with ischemic stroke.the polymorphisms of C1236 T ? G2677T/A in the ABCB1 gene have no relationship with AR.The haplotypes(C-T-T,T-G-C,T-T-T)may increase the risk of AR in ischemic stroke patients.
