Objective: The effect of histone deacetylase(HDAC)inhibitors as anti-tumor therapies has been evaluated.The purpose of this study was to evaluate the potential of Yin-Yang1(YY1)governs the sensitivity of HCC cells to HDAC inhibitors.Materials and Methods: We evaluated the effect of the HDAC inhibitors on 10 HCC related cancer cell lines using CCK-8 assay.We performed YY1 gene expression govern the the sensitivity to HDAC inhibitor in HCC,including knockdown and over-expression.We demonstrated HDAC inhibitors treatment effect on HCC may be affected by the expression of YY1.Results: According to the Ward's Cluster analysis,we evaluated the high and low sensitivity of HDAC inhibitors on HCC related cancer cell lines.Using gene expression profiling,we identified YY1 mediates HDAC1 expression to reduce the sensitivity to HDAC inhibitor in HCC.Conclusions: Our study highlight YY1 in conjunction with HDAC1 as a key regulator of the cellular response to HDAC inhibitor in HCC.Hence,YY1 may be a potential therapeutic target to reverse sensitivity to HDAC inhibitors in HCC.
