Objective:Oral squamous cell carcinoma(OSCC)is attributed to smoking,alcohol abuse and long-term physical and chemical stimulation.But a large number of clinical practice showed that the occurrence rate of no obvious physical and chemical stimulation cancers significantly increased and Human papillomavirus(HPV)was pathogenic factor to OSCC.The study is to show a detection about the expression of telomerase in human oral mucosal epithelial cells which was infected with HPV,make clinical detection to HPV infection of OSCC and explore the relationship between OSCC and HPV infection.Materials and Methods:1.Normal human oral epithelial cells were cultured in vitro and infected with HPV16 E6.The expression of telomerase was detected respectively which were from normal epithelial cells,HPV16 E6 infected epithelial cells,HPV16 E6/E7 immortalized epithelial cells and Tca8113 tongue squamous cell carcinoma cells.2.The specimens of 53 patients with OSCC were collected,including tumor tissues,adjacent tissues and normal oral mucosa.The normal oral mucosa of the patients with non OSCC in 100 cases were taken as control group.PCR-DNA reverse dot hybridization method of HPV genotyping detection technology was used to detect the infection of HPV in every group.Results:1.Normal human oral mucosal epithelial cells were not detected in telomerase activity,HPV16 E6 transfected oral mucosal epithelial cells telomerase is activated,the telomerase activity in HPV16 immortalized oral epithelial cells was approximately 4times of HPV16 E6 transfected oral epithelial cells(P<0.05).The telomerase activity of tongue squamous cell carcinoma cells was approximately 8 times higher than oral epithelial cells by HPV16 E6(P<0.05),at the same time,was significantly higher than that of HPV E6/E7 immortalized epithelial cells.2.Eight in 53 cases with OSCC were HPV positive,the infection rate 15%,including 1 case with HPV mixed infection(HPV high-risk type and low-risk type)and 7 cases with HPV single infection(only HPV high-risk type).1 case with HPV mixed infection by high-risk type and low-risk type was detected in the control group,the infection rate 1%.There was significant difference in HPV infection rate between the experimental group and the control group,P<0.05.3.8HPV positive OSCC patients,4 cases in stage ???,4 cases in stage ???,45 HPV negative OSCC patients,25 cases in stage ???,20 cases in ???.There was no significant difference between two groups,P>0.05.4.8 HPV positive OSCC patients,3cases has cervical lymph node metastasis,5 cases has no cervical lymph node metastasis,45 HPV negative OSCC patients,20 cases has cervical lymph node metastasis,25 cases has no metastasis.There was no significant difference between two groups,P>0.05.Conclusions:1.HPV may play a regulatory role in oral epithelial cells cancer 2.HPV is closely related to the occurrence of oral squamous cell carcinoma.3.Incidence of HPV infection in cases with OSCC was significantly higher than those no OSCC.But HPV infection with OSCC had no impact on the clinical stages and lymph node metastasis.
