Objective: To use human circRNA microarray to screen the differential expression of circRNA of plasma between hypertensive patients and normal controls and to study of circRNA expression profile.Use real-time quantitative polymerase chain reaction(RT-PCR)technology to verify the expression of circRNAs and predict the potential pathogenesis through bioinformatics analysis,which lay the foundation for the further studies on whether circRNA can act as diagnosis biomarkers of hypertension and the potential therapeutic targets.Methods: We chose 5 cases of hypertension and 5 cases of normal controls as the participants in this research.Peripheral venous blood from each participant was collected to separate the plasma,which the total RNA was extracted from.Human circRNA microarray was performed to detect the difference of circRNA expression between the two groups.Through the data analysis,we get the expression profile of circRNA.Then,we used q RT-PCR technology to validate the expression of the circRNAs between 49 cases of hypertensive patients and 49 cases of normal controls.Finally,we predicted miRNAs associated with the circRNA and the circRNA targets and then Gene ontology analysis and KEGG pathway analysis were applied for further enrichment of mRNA data.Results: Compared with the normal group,a total of 59 circRNAs were differentially expressed in hypertension group,including 46 up-regulated and 13 down-regulated.Two up-regulated circRNAs(hsa-circ-0000437,hsa-circ-0008139)and two down-regulated circRNAs(hsa-circ-0005870,hsa-circ-0040809)were validated by RT-PCR.The results showed that hsa-circ-0005870 was down-regulated,consistent with the microarray data.While,the expression levels of hsa-circ-0000437,hsa-circ-0008139 and hsa-circ-0040809 were no significant differences between two groups.The result of bioinformatics indicated that there were 5 miRNAs and 849 corresponding mRNAs.GO analysis showed the targets of hsa-circ-0005870 may involve in biological process such as RNA polymerase activity and KEGG analysis showed the main pathway of the targets is TGF-? signal pathway.Conclusion: We obtained the circRNA expression profile in hypertension and verified that the expression of hsa-circ-0005870 in hypertension group is down-regulated.The bioinformatics analysis indicated that hsa-ciightrc-0005870 might regulate the development of hypertension through TGF-? signal pathway.The study lays foundation for circRNAs as diagnosis biomarkers and potential therapeutic targets and provides a new direction for the research of the pathogenesis of hypertension.
