| Objective: Through the detection of the expression of TLR4 and NF-K B in gastric cancer and adjacent tissues,and explore their relationship with clinical pathology and prognosis in patients with gastric cancer,analysis the follow-up data to evaluate TLR4 and NF-K B in the course of the occurrence development and prognosis in patients with gastric cancer..Methods:(1)We collect 122 cases with gastric cancer and 33 adjacent normal tissues from the First Affiliated Hospital of Shihezi University Medical College(122 cases of gastric cancer patients had completed data,and have been followed up).(2)122 patients were followed up once a year(telephone,visit)and the last follow-up time is October 1,2014,the longest follow-up time has exceeded 5 years.(3)The gastric cancer and adjacent tissue paraffin block were made into tissue microarrays,the levels of TLR4 and NF-K B expression was detected by immunohistochemistry in tissue microarray.(4)Data was analyzed using SPSS19.0 software,statistical analysis use non parametric method,Kaplan-Meier method test,survival analysis,Cox regression analysis and Spearman correlation analysis.Results:(1)In gastric cancer tissues,the positive expression rate of TLR4 in gastric carcinoma was 92.6%(113/122),and the negative expression rate was 7.4%(9/122).And in adjacent tissue,the positive expression rate was about 72.7%(24/33),and the negative expression rate was 27.3%(9/33).(2)TLR4 and NF-K B expression in gastric carcinoma were higher than the adjacent normal tissue(P<0.001).(3)In gastric cancer tissues,the positive expression rate of NF-ΚB in gastric carcinoma was 96.7%(118/122),and the negative expression rate was 3.3%(4/122).And in adjacent tissue,the positive expression rate was about 57.8%(25/33),and the negative expression rate was 42.2%(8/33).(4)The expression of TLR4 was correlated with gender,clinical stage,invasion depth and lymph node metastasis,and the percentage of TLR4 positive cells in tumor adjacent tissues was negatively correlated with the survival time.(5)The Cox regression analysis showed that the pathological type,age,differentiation,lymph node metastasis,distant metastasis and the expression of NF-kappa B were independent prognostic factors of gastric cancer patients(P<0.05).Conclusions:(1)In gastric carcinoma,the expression of TLR4 and NF-ΚB were higher than in adjacent tissues.(2)The high expression of TLR4 in gastric cancer tissues was correlated with depth of invasion,lymph node metastasis and clinical staging in patients.And the high expression of NF-kappa B was associated with prognosis in patients with gastric cancer,suggesting that TLR4 and NF-kappa B take part in the growth and transfer of gastric cancer tissue.(3)The optimal combination of predicting the prognosis of gastric cancer patients is summarized. |
