Object The expression levels of PTEN and mi RNA-21 were detected by real-time fluorescence quantitative PCR(q RT-PCR)to observe the differences of PTEN and mi RNA-21 expression levels between Xinjiang Han type 2 diabetes mellitus(T2DM)patients and normal glucose tolerance(NGT)people,and to investigate the correlation of PTEN gene and mi RNA-21 in Xinjiang Han T2DM.Methods50 cases of Xinjiang Han T2DM patients were selected as experimental group(T2DM group)while50 subjects with NGT as control(NGT group).The general data including gender,age,height,weight,waist circumference,hip circumference and blood pressure were measured and recorded,and then calculated the body mess index(BMI)and waist-to-hip ratio(WHR).Detected the fasting plasma glucose(FPG),2-hour postprandial plasma glucose(2h PG),fasting plasma insulin(FINS),fasting C peptide,glycosylated hemoglobin(Hb A1c),triglyceride(TG),total cholesterol(TC),high density lipoproteincholesterol(HDL-C)and low density lipoprotein-cholesterol(LDL-C),and then calculated the HOMAinsulin resistance index(HOMA-IR).The expression levels of PTEN gene and mi RNA-21 were detected by the q RT-PCR.All the statistical analyses were performed by the SPSS 20.0 software.Results1.Compared with NGT group,the BMI and WHR of Xinjiang Han T2DM group were significantly higher(P<0.05).There was no statistically significant difference in gender,age,SBP and DBP between the Xinjiang Han T2DM group and the NGT group(P>0.05).2.Compared with NGT group,the levels of FPG,2h PG,Hb A1 c,In(HOMA-IR),TG,TC and LDL-C in Xinjiang Han T2DM group were higher,the levels of FINS,fasting C peptide and HDL-C were lower,the differences were statistically significant(P<0.05).3.The expression level of PTEN in Xinjiang Han T2DM group showed 2.18 times than NGT group.4.The expression level of mi RNA-21 in Xinjiang Han T2DM group showed 2.12 times than NGT group.5.There was no significant correlation between the expression levels of PTEN gene and mi RNA-21 in Xinjiang Han T2DM group(r=-0.078,P=0.592).Conclusion1.The higher expression levels of PTEN and mi RNA-21 in T2DM may be involved in the occurrence and development of T2DM.2.There may be no correlation between the expression of PTEN gene and mi RNA-21 in Xinjiang Han T2DM.
