?backgroud?Pancreatic cancer is a highly malignant tumor in the digestive tract system.Pancreatic cancer is characterized by its insidious onset,low early diagnosis rate and high mortality",known as the "king of cancer." Early diagnosis is the best way to improve the prognosis of pancreatic cancer,but it has no specific and high sensitive biological markers.Based on this,our group used the mass spectrometry combined with label-free technique to analyze the protein expression of three fresh pancreatic cancer tissues and adjacent tissues,and obtained the Chymotrypsin C that the specific expression protein of pancreas.CTRC is one of the members of the serine protease family.It produced by pancreatic acinar cells and secreted by pancreatic ductal cells.CTRC's main function is that to participate in the activation of trypsinogen and degradation of trypsin to maintain the integrity of pancreatic tissue structure and function.Additionly,it participates in the regulation of intestinal and blood calcium concentration to play a physiological role.There is little research about CTRC and pancreatic cancer.At present,research find the expression of CTRC is down regulated in pancreatic cancer,and is related to the metastasis of pancreatic cancer,but the specific mechanism is unknown.Autophagy is closely related to tumor.In the early stage of cancer,autophagy can inhibit the growth and metastasis of tumor by inhibiting cell proliferation and increasing apoptosis.In the advanced and late stages of cancer,autophagy promotes tumor growth and metastasis by enhancing the adaptation of tumor cells to the environment.Autophagy can inhibit or promote the occurrence and development of pancreatic cancer,and can be used as a target for clinical treatment.To sum up,CTRC and autophagy are closely related to pancreatic cancer,but what is the mechanism of the two in pancreatic cancer? Is there a connection between the two? ?Objective?1.To clarify the difference of the expression of CTRC in pancreatic cancer tissues and cancer adjacent tissues.Discussing the relationship between the expression of CTRC and prognostic factors in patients with pancreatic cancer and analysising the clinical significance.2.To clarify the relationship between the expression of CTRC and autophagy related protein LC3,and provide the experimental and theoretical basis for the study of pancreatic cancer.?method?The expression of CTRC and LC3 was detected by immunohistochemical staining in 180 pancreatic cancer tissues and adjacent tissues.Wilcoxon rank sum test analysis of expression of carcinoma and adjacent tissues;Sperman correlation analysis of CTRC and LC3 expression levels with prognosis of patients and correlation of CTRC and LC3;Kaplan-Meier survival curve and COX regression model analysis the relationship between the expression of pancreatic cancer and the survival of the patients with CTRC and LC3;the receiver operating curve to evaluate the accuracy of CTRC immunohistochemical staining in the diagnosis of pancreatic cancer.All statistical analysis was done using SPSS19.0 statistical software,and P<0.05 indicated that the difference was statistically significant.?results?1.The expression of CTRC in pancreatic cancer and paracancerous tissues was significantly different(P<0.001).The expression of CTRC in pancreatic cancer was related to T stage(P=0.014),tumor site(P=0.018)and clinical stage(P=0.013).There was no significant correlation between CTRC expression and survival(P=0.7287).Analysis of the receiver operating curve: the area under CTRC of ROC was 0.921(95%CI 0.883~0.959).2.The expression of LC3 in pancreatic cancer and paracancerous tissues was significantly different(P<0.001).The expression of LC3 was significantly correlated with the survival time(P=0.0151),and the higher the expression,the worse the prognosis.And the risk of pancreatic cancer in LC3 positive expression was negative 1.89 times.The expression of CTRC was significantly correlated with LC3(r=0.581,P< 0.001).?Conclusion?The expression of CTRC was significantly down regulated in pancreatic cancer.Further analysis showed that CTRC was correlated with T stage,tumor site and clinical stage.At the same time,the diagnostic efficacy of CTRC is very high.CTRC can be used as a therapeutic target for clinical application,and it can provide a basis for clinical pathological diagnosis.CTRC is associated with LC3,and LC3 is an autophagy related protein,which indirectly suggests that CTRC may be related to autophagy,which provides a new way to further study the pathogenic mechanism of CTRC.
