| Background: Prostate cancer is one of the most common cancers of male urinary system,affecting the health of men in Worldwide.It is the second most common tumor which leading death of male.In 2012,there is account for 15 percent of new tumor patients of prostate cancer.In 2017,there are about 161360 cases of new patients and 26730 deaths in United States and have a tendency to increase.With the aging of the population,the burden of prostate cancer is expected to increase significantly in the next few years.For the treatments of prostate cancer include of watchful waiting,active surveillance,radical prostatectomy,hormonal therapy,radiotherapy and chemotherapy,etc.The therapeutic effect is poor for castration resistant prostate cancer,and most of castration resistant prostate cancer will shift to other site,and the mortality rate is about 50 percent.Therefore,we urgently need a kind of model that can simulate the characteristics of prostate cancer,in order to research mechanism of prostate cancer and its treatment better,and build platform for further research.Objective:1.Using nude mice to establish patient-derived xenograft models of prostate cancer;2.To identify histology and molecular characteristics of model and compare with of the original tumor;3.To establish an experimental platform for further drug tests.Methods: 1.Collect fresh specimens of 44 cases of prostate cancer and build P1 generation of PDTX by subcutaneous transplantation;2.Establish P2,P3 Models of PDTX;3.Collect of patients’ clinical data and statistical analysis various factors that affecting graft,providing data for future;4.Detection recovery ability of tumor after frozen;5.Identification of model and compare with original tumor tissue of the characteristics.Results: 1.Built 3 cases of PDTX of prostate cancer,and the rate of success model is 6.8 percent.Age of patient has nothing to do with the rate of model(P = 0.259).Gleason scores of prostate cancer have nothing to do with the rate of model(P = 0.650).PSA The value of PSA has nothing to do with the rate of model(P = 0.397).The metastasis of tumor is benefit for grafting(P = 0.045).2.The rate of tumors recovery after frozen is 100 percent,and the grafting rate of P2 and P3 generation are 100 percent.3.The structure and characteristics of model tumors are similar with the original tumor.4.The model tumors have the characteristics of human prostate cancer cells by STR test.Conclusions: 1.The method of establish PDTX model is reliable,and model tumors maintain the heterogeneity of original tumor.2.The PDTX models which are established in this research can be used in further experiments of prostate cancer. |
