Study On Autophagy And Apoptosis Of PC12 Cells Induced By Serum Starvation,RTS And Arsenic

The pathogenesis and treatment of neurodegenerative diseases is a problem that the scientists have been trying to overcome,and its regulation by autophagy has been become one of the research hotspots.Autophagy is a highly conserved general physiological phenomenon in eukaryotic cells,which can both degrade cytopathic proteins and organelles to maintain the stability of the cell environment,but also change the fate of cells under stress conditions.This study focus on the changes of autophagy and apoptotic in neuronal cells under serum starvation,RTS and arsenic ions at different valences.In this paper,PC12 cells induced by NGF were used as experimental model.Experiments show that: the autophagy of PC12 cells increased within serum starvation 3-12 h,and decreased within 18-24h;at the same time the apoptosis of PC12 cells did not change significantly within serum starvation 3-18 h,but increased 24 h.The results show the serum starvation conditions can induce protective autophagy in PC12 cells.The levels of autophagy in PC12 cells were different under realgar,ATO,sodium arsenate and RTS.In the case of realgar and ATO treatment,the level of autophagy showed a dose-dependent change,which were highest at 0.1?g/ml.The change of autophagy for PC12 cells increases initially and decreases afterwards,and hightest at thw arsenic concentration reached at 1ug/ ml.In addition,the apoptosis of PC12 cells increased exposured in realgar,ATO,sodium arsenate and RTS.The results show the serum starvation and four arsenic agents can induce protective autophagy in PC12 cells.Among the four arsenic agents,the effect of RTS on apoptosis was minimal.Indicating that the cytotoxicity is reduced in PC12 cells change realgar into RTS,suggesting that RTS has the potential to replace realgar in the future.
