Arbovirus which spread by mosquito biting can lead to the outbreak of many viral vector borne diseases.More than 500 kinds of arboviruses have been found in the world.The mosquito borne arboviruses account for more than 50% of the total.Shanghai is one of the largest ports in the world,contributed to the diversity and complexity of biological vector species.Through a survery of arboviruses carried by mosquitoes and midges provide a theoretical basis for the prevention and control of arboviral diseases in Shanghai.We isolated the mosquito samples by cell separation for the virus.Five strains of virus were isolated.Of which,two strains of Japanese encephalitis virus were further analyzed for biological characteristics and molecular characteristics.At the same time,establish one step TaqMan probe method to identify the virulent strains of Japanese encephalitis virus.The results show that,two isolates of the five viruses were Japanese encephalitis viruses and the other three isolates were banna viruses.Two strains of Japanese encephalitis virus were found to be different from those found in the amino acid residues of E protein.Intracerebral injection can cause disease in mice.A method of one step TaqMan probe method was established successfully for identification of virulent strains of Japanese encephalitis virus.
