| In recent years,Social security issues caused by drug abuse have been in an increasing tendency,which endanger the personal safety of the patient.How to effectively detect and monitor drugs,prevent the illegal circulate has become an important facing issue for our country.Although some existing detection methods for drug abuse can detect accurately,but they have some problems such as low efficiency,difficult to immediately detect special drugs,and sometimes cost high.Surface-enhanced Raman spectroscopy(SERS)technique,an emerging analysis technology,has high sensitivity and low water-disturbance,etc.Since the late 1900 s,it has been widely used in many fields like food security,drug detection and material analysis,etc.,and has a big value for Rapid real-time detection of drug abuse.In this paper,SERS substrates have been prepared and characterized,the best conditions of SERS detection for three controlled substances was explored,and the works of detection and semi-quantitative analysis has been accomplished,which are of a certain meaning.Firstly,based on the density functional theory(DFT),utilizing the B3 LYP level of Gaussian software,the geometry optimization and spectra calculation for Scopolamine hydrobromide,Promethazine hydrochloride,atropine sulphate have been obtained.The results of theoretical analysis were compared with experimental results,and the Raman frequencies and their vibrational modes were assigned according to the visual software.These jobs are prepared for the follow-up experiment.Then,the silver colloid SERS was prepared by chemical reduction method,and the properties of the colloid are characterized by field emission scanning electron microscopy(SEM)and Dynamic light scattering device.The silver colloid has uniform size,and a good batch to batch reproducibility.In addition,a highly hydrophobic and reflectivity substrate was investigated to use in SERS detection.Finally,a simple and rapid SERS method to detect Scopolamine hydrobromide,Promethazine hydrochloride,atropine sulphate was established based on silver colloid.Drug in liquid samples were detected.The SERS enhancement effects of silver nanoparticle induced by different aggregation agents were researched,and the aggregation agents with the best results were determined,respectively.The limit of detection(LOD)are 0.1μg/m L,0.61μg/m L,0.5μg/m L and the recovery rates are between 98.5%~109.7%,96.5%~108.8%,98.3%~107.4%,respectively.This method is simple,quick and has a great practical potential for the detection of abuse drugs. |
