Objective: To explored whether can the intervention of Stauntonia chinensis(SC)reduce the damage,which caused by long-term exposure to cadmium,of liver and kidney.Methods:There were 50 male SD rats stratified by weight and divided into 5 groupes(10 in each group)randomly,including control group(Control,saline,by gavage),the cadmium group(Cd,saline,by gavage),and three treatment groups: SC low-dose[SCL(0.5 g/kg),by gavage],SC median-dose [SCM(1 g/kg),by gavage],SC high-dose [SCH(2 g/kg),by gavage].Cd Cl2(50mg/L)were added in drinking water of rats,except the control group.All rats were sacrificed after 90 days feeding.Blood sample and organ sample including liver and kidney were collected after anesthetization and stored in-80 refrigerator.The cadmium content of liver,kidney and ultrastructure were tested by Graphite Furnace Atomic Absorption Spectrometry,liver and kidney were examined by histopathology and electron microscopy.MDA、SOD、GSH-Px were measured using kit.Mt-1 m RNA、Mt-2 m RNA expression were analyzed by fluorescence-based real-time PCR.Western blot was adopted to analyse the expression of MT.Results: The organ coefficients of liver and kidney in the Cd group was higher than that in the control group(P < 0.05),the organ coefficients in SCH group were significantly reduced(P < 0.05).Compared with the Cd group,AST,ALT,creatinine and urea were reduced in other groups(P<0.05).Liver histopathological observation showed hepatocytes is swelling,pyknosis and necrosis,inflammatory cells were seen,SC intervention can improve the liver injury,inflammation and reduce hepatocytes necrosis.Kidney histopathological observation showed that the arrangement of renal tubular is orderly,the lumen is smaller and the tubular epithelium cell is swelling and necrosis.SC intervention can improve the kidney injury,and the swelling of the tubular epithelium cell.In Cd group,electron microscopy showed that glomeruli podocyte foot secondary processes is uneven distribution,thickening of the glomerular basemen membrane.Hepatic ultrastructure showed mitochondria and endoplasmic reticulum are swelling and disappear.SC intervention can improve the ultrastructural damage of liver and kidney.Compared with the Cd group,the cadmium contents of liver,kidney,mitochondria and microsome were reduced in SC intervention group(P < 0.05),with the cadmium contents of cytoplasm were increased(P<0.05).In Cd group,liver and kidney showed high levels of MDA,and decreased levels of SOD、GSH-Px(P<0.05).Compared with Cd group,liver and kidney showed decreasing levels of MDA and increaseing levels of SOD、GSH-Px in SCM group and SCH group(P<0.05).In SC treatment groups,the gene expression of Mt-1,Mt-2 were higher than Cd group(P<0.05).Western blot analysis of MT levels revealed that SCM group and SCH group increased MT protein levels,and were higher than Cd group(P<0.05).Conclusion: Stauntonia chinensis may relieve the injury of liver and kidney of were long-term cadmium exposed rats.On one hand,Stauntonia chinensis may increase the activitie of antioxidant and relieve Cd-induced oxidative damage;on the other hand,Stauntonia chinensis may increase MT expression to bind to the free Cd that relieve Cd-induced organ toxicity.
