Objective: To investigate the correlation between the level of mi R-125 and EGFR-TKIs resistance and its predictive value in non-small-cell lung cancer by detecting the levels of mi R-125 in serum before and after EGFR-TKI resistance.Methods: Real-time quantitative PCR was used to detect the expression of levels of mi R-125a-5p and mi R-125 b in the serum samples collected from 36 patients without EGFR-TKIs resistance,20 patients with EGFR-TKIs resistance,and 13 patients with benign lung disease.We followed up and evaluated the short-term efficacy of NSCLC patients with EGFR-TKIs.Results:1.The expression levels of mi R-125a-5p in the serum of EGFR-TKIs non-resistant group 1.058(0.506 ~2.313)and resistant group 0.603(0.286 ~ 1.154)were significantly lower than those of benign lung disease group 2.174(1.655~3.317)(P=0.014,P=0.000).The expression of mi R-125a-5p in the resistant group was significantly lower when compared with the non-resistant group,and the difference was statistically significant(P=0.045).However,the levels of mi R-125 b were not statistically significantly different among three groups(P > 0.05),and the difference between non-resistance group1.025(0.441 ~ 2.957)and resistance group 1.684(1.021 ~ 2.532)also did not reach statistical significance(P=0.259).It has been found that the levels of mi R-125a-5p and mi R-125 b decreased after drug resistance in 3 paired serum samples.2..The levels of mi R-125 were no statistically significantly different in T790 M mutation group and no T790 M mutation group in the serum of patients with EGFR-TKIs resistance(P=0.877,P=0.280).3.The expression of mi R-125a-5p and mi R-125 b in the treatment failure group was lower than that in the beneficial group,but had no significant difference(P=0.290,P=0.264).Conclusion:1.The low levels of mi R-125a-5p in serum may be related to the EGFR-TKI acquired resistance of NSCLC.mi R-125a-5p may be not only a potential marker of EGFR-TKI resistance,but also an effective index for predicting the efficacy of EGFR-TKIs.2.The expression of mi R-125 were no statistically significantly different in T790 M mutation group and no T790 M mutation group in the serum of patients with EGFR-TKIs resistance...
