ObjectiveThe study aimed to detect the differently expressed proteins in serum of polypoidal choroidal vasculopathy(PCV),using protein chip technology,to find the early diagnostic marker and drug targets for PCV and provide evidence for further exploring the pathogenesis of PCV.Methods1.One case of negative control,one case of AMD and two cases of PCV serum samples were detected by AAH-BLG-507 protein chip,and analysis of differentially expressed proteins were identified.2.Thirty-four kinds of cytokines and chemokines were selected to compose a multiplex immunoassay kit according to the results of AAH-BLG-507 protein chip.Serum samples form 65 cases of PCV(43 males and 23 females,age ranged from 49 to 81 years old)and 70 control individuals(40 males and 30 females,age ranged from 53 to 78 years old)were analyzed by the multiplex immunoassay kit.The different expressions of the factors between the two groups were evaluated.Results1.The average age of PCV was(65.43 ± 8.57)years old,which had no significant difference compared with the control group(P=0.07>0.05).The gender also had no statistical difference between the two groups(P>0.05).2.The results of protein chip test shows that 79 proteins were differentially expressed in PCV,including 58 proteins up-regulated and 21 proteins down-regulated.The differentially expressed proteins were consisted of cytokines,chemokines,extracellular matrix proteins/matrix metalloproteinases,growth factors and their receptors.3.The results of the multiplex immunoassay kit show that 10 proteins weredifferentially expressed in PCV.The average concentrations of Eotaxin(52.618±24.954)pg/mL,IL-1?(0.481±0.279)pg/mL,IL-1RA(289.232±146.678)pg/mL,IL-4(2.069±1.050)pg/mL,IL-5(0.169±0.111)pg/mL,IL-13(0.424±0.122)pg/mL,IL-22(4.730±3.790)pg/mL,GM-CSF(1.542±0.752)in PCV were higher than that in the control group(P value of each pair were 0.003,0.007,0.009,0.002,0.040,0.003,0.003,0.003).The average concentrations of IL-7(2.929±2.749 pg/mL)and SDF-1?(347.141±101.243pg/mL)were lower than that in the control group(P value were 0.003 and 0.008,respectively).Conclusion1.The protein chip technology is a rapid,simple high-throughput proteomics research methods,provides a new research platform for the research of PCV.2.PCV may be related to the high expression of Eotaxin,IL-1RA,GM-CSF,IL-1?,IL-4,IL-5,IL-13,IL-22 and the low expression of IL-7,the SDF-1?.3.The different expression proteins may provide biological markers for early diagnosis and targeted drug treatment factors of PCV.
