increased year by year.Due to the traditional use of antifungal drugs and candida albicans biofilm formation,its drug resistance to gradually strengthen,drug resistance mechanism is more and more complex,thus increasing the treatment difficulty of biofilm related infections.The effective treatment and does not cause related viscera damage of antifungal drugs is extremely limited.So a growing number of researchers trying to find some new antifungal drugs from Chinese herbal medicine used in clinical,or with the traditional joint application of antifungal drugs,set up in addition to the traditional antifungal drug treatment of another world.Tannin exist widely in plant kingdom,the most of the crude drug contains tannin compounds,it has antiviral effect,antibacterial effect,antitumor and anticancer effect etc,and the side effects of small,easy to extract.Persimmon is rich in tannins,has been widely cultivated in china,the production is rich.Therefore this study using the ethanol ultrasonic extraction and the persimmon tannin extract,using XTT reduction colorimetric method,observe the persimmon tannin water extract on candida albicans biofilm in vitro inhibitory effect,to provide experimental basis for clinical treatment of candida albicans disease.Objective:Candida albicans in vitro model of biofilm on the antifungal drug sensitivity test found that the strains of candida albicans that exist in the form of biofilms are highly resistant to conventional medicine.Therefore,clinical treatment of candida albicans infection is difficult.Tannins can be solidified microbe protoplasm and used in a variety of enzymes,to a variety of bacteria have obvious inhibitory effect.This experiment studied the persimmon tannin water extract on candida albicans biofilm in vitro inhibitory effect,persimmon tannin extract water clear effect on candida albicans biofilm.Methods:1 Persimmon tannin extracton the persimmon Gai fruit smash all grinding,soaking in ethanol after ultrasonic extraction,adsorption in addition to sugar,rotary evaporation concentrated solution with distilled water and diluted with AB-8 macroporous resin,polysulfone hollow fiber ultrafiltration membrane ultrafiltration,the molecular weight of persimmon tannin solution 10000,persimmon tannin content determination of water extract liquid tannin using Folin-Denis method.Take full persimmon tannin extract water,use 0.22 ?m bacteria filter to filter in addition to bacteria,4 avoid light preservation.Set aside.?2 The preparation of candida albicans biofilms:Candida albicans administered in YPD medium,30 ?shaking bed for the night train,Centrifugal collecting bacteria,PBS cleaning bacteria 2 ~ 3 times,Match the concentration of 1x106cfu· mL-1 standard bacteria liquid with RPMI-1640 Medium.Will 100 ?L standard microbial inoculation in 96-well culture plate,Plus RPMI-1640 medium volume up to 200 ?L,37 ?stand for 24 h,Take shape Biological membrane structure.Observed under inverted microscope candida albicans biofilm formation.3 Persimmon tannin extract water effect on candida albicans biofilm:Persimmon tannin XTT reduction colorimetric method are used to observe water extract of candida albicans membrane,Evaluation of different concentrations of persimmon tannin extract water of different growth stages of drug susceptibility of candida albicans,use Inverted microscope to observe the drug after candida albicans biofilm morphology.Results:1 The tannin content of Persimmon tannin extract water is 7.42 mg/mL.2 Persimmon tannin extract water on candida albicans film formation and the growth of hypha cells has obvious inhibitory effect(P<0.05),Its function and drug concentration were positively correlated,Of 3.7100 mg/mL and 1.8550 mg/mL persimmon tannin extract water on candida albicans biofilm formation inhibition rate of more than 90%.3 Persimmon tannin extract water sensitivity to different growth stage of candida albicans drugs is not the same,The high concentrations of persimmon tannin extract water(3.7100 mg/mL and 1.8550 mg/mL)on the growth stage of candida albicans biofilm inhibition rate greater than 90%,the middle concentration and the low concentration pesimmon tannin extract water(0.9275 mg/mL,0.4638 mg/mL,0.2319 mg/mL 0.1159 mg/mL)under the condition of the same concentration,along with candida albicans biofilm maturation,persimmon tannin on the inhibition rate of the decline(P<0.05).Conclusions:1 Persimmon tannin water extract has obvious inhibitory effect on candida albicans biofilm formation and the growth of hypha sells(P<0.05),and positively correlated with drug concentration.2 Drug susceptibility of persimmon tannin water extract on different time of candida albicans is not conformity.Low concentration of persimmon tannins in the same concentration,as the candida albicans biofilm matures,inhibition rate of persimmon tannin water extract on candida albicans has been declining.
