Polygonatum sibiricum Red.is the rhizome of Polygonatum kingianum Coll.et Hemsl.,Polygonatum sibiricum Red.,Polygonatum cyrtonema Hua.for liliaceous plant which is sweet,natured and can restore vital energy,tonify spleen,lung and kidney.As the commonly traditional Chinese medicine,Polygonatum sibiricum Red.belongs to the "medicine food homology" plants,which is a perennial herbaceous plant.The rhizomes of Polygonatum sibiricum Red.contain Polygonatum sibiricum Red.polysaccharide,steroidal saponins,anthraquinone compounds,alkaloids,lignans,vitamins,and amino acids used for human body,which can make medication.Polygonatum sibiricum Red.is a perennial herbaceous plant and poor growth,whose rhizomes elongation grows one section each year.At present,a lot of Polygonatum sibiricum Red.is wild,and the harvest year is appropriate or not,which will directly affect the quality and efficacy of the medicinal materials.So this research mainly aim at different growth period in Polygonatum sibiricum Red.chemical composition studied,and compare each component content in different growth period and the influence of different drying methods on the content determination for Polygonatum sibiricum Red.reasonable harvest time and providing the theory basis for Chinese medicine quality control and reference.Using the 3,5-dinitrosalicylic acid method combined with anthracene ketone-sulfuric acid method for determination of rhizoma polygonati polysaccharide,using response surface analysis of anthracene ketone-sulfuric acid colorimetry determination conditions were optimized,the best determination conditions are obtained.Establish isothiocyanate benzene ester(PITC)column before derivative HPLC on yellow liquor for 16 kinds of amino acids in the determination of using statistical software SPSS correlation analysis to it.Using high performance liquid chromatography(HPLC)-dual wavelength method to simultaneous determination of five active chemical constituents in rhizoma polygonati content,and compare each component content in different growth period,for the yellow fine reasonable harvest time and quality control of Chinese traditional medicine to provide theoretical basis and reference.Specific results are as follows:(1)In the anthrone-sulfuric acid method,three experimental conditions including concentrated sulfuric acid mass fraction,anthrone dosage and color development time were optimized by single factor experiments and response surface methodology using absorbance value as the dependent variable.The method combined with 3,5-dinitrosalicylic acid method to determine the reducing sugar content of Polygonatum Sibiricum Red.and compared the polysaccharide content in different years.Result: The optimum experimental conditions were found as follows: 87% concentrated sulfuric acid,0.2g of anthrone dosage and 25 min of color development.The total sugar and reducing sugar content of Polygonatum sibiricum Red.of four year is the highest in test.(2)The result indicated that the correlation coefficient square was above 0.9983 in the linear range of the 16 kinds of amino acids.The limit of determination was 0.38~0.92?mol/L.The limit of quantification was 0.95~ 2.31?mol/L.The mean spiked recovery rate of the sample was 83.20%-99.09%,with a relative standard deviation of 0.26%-2.68%.Vacuum freeze drying is better than other drying methods.The content of amino acids of growing period of 4 years is higher than others.Moreover,the essential amino-acid content was positively correlated with total amino acids content(p<0.01),and there were certain positive correlation among different kinds of amino acids.(3)The extraction conditions of five chemical constituents(adenosine,ginsenoside Rb1,?-sitosterol,vanillic acid,liquiritigenin)were optimized as follows:6.0g sample and 40 m L 70 percent of methyl alcohol were added in the conical flask,then ultrasonicated for 30 min,and subsequently the extractive was analyzed by UV-detector at 252 nm and 280 nm,reversed phase column Diamonsil C18(4.6 mm×250 mm,5 ?m),and 0.1% aqueous formic acid-0.1% acetonitrile formic acid as the mobile phase with flow rate at 1.0 m L/min.The Analysis was performed at 35?.The linear calibration ranges of five chemical constituents were within 0.5~100?g/m L with good linearity(R2>0.999).The limit of determination was 0.3~0.5mg/kg.The limit of quantification was 1.1~1.6mg/kg.The mean spiked recovery rate of the sample was 97.2%~101.2%,with a relative standard deviation of 1.4%~2.9%.Through the above research growing period of four years of Guizhou Polygonatum sibiricum Red.in the chemical composition content is relatively high,and vacuum freeze drying way is better than other drying effect.As a result,.it can be drawn from the Polygonatum sibiricum Red.best harvest year for three years after planting,and the best dry way of fresh Polygonatum sibiricum Red.is vacuum freeze drying.These results to build Guizhou Polygonatum sibiricum Red.processing of the normative production provides an important gist,offering the research data to clarify the Polygonatum sibiricum Red.development and utilization of the medicinal value.
