Abstract/Summary:
Objective: To explore the influence of hypoxia on the phagocytosis and killing functions,expressing surface molecules and secreting cytokines of peritoneal macrophages in mice by establishing model under the plateau hypoxia environment.Methods: After exposing mice to an altitude of 400 m,2200 m and 4200 m for 30 days,(1)Using flow cytometry to detect the phagocytosis and killing functions of peritoneal macrophages can be loaded of staphylococcus aureus to FITC;(2)Detecting the respiratory burst level of the cultured macrophage supernatant in mice with2′,7′-Dichlorodihydrofluorescein diacetate(DCFH-DA)fluorescent probe method.(3)Detecting the concentration of NO stable oxide metabolites nitrite(NO2-)of the cultured macrophage supernatant in mice with ELISA kits.(4)Using flow cytometry to detect the changes of peritoneal Mφ surface molecules.(4)Detecting the concentration of IL-6 and TNF-α of the cultured macrophage supernatant in mice with ELISA kits.Results: In comparison with the 400 m group,Mφ to gobble up ability,breathing capacity and NO release a quantity to reduce;Mφ CD14、CD64、TLR4、CD11b、CD18、CD80、MHC-Ⅱ in mice were decreased markedly(P<0.05)after exposed to hypoxia at 4200 m and 2200 m for 30 days.In addition,in comparison with the2200 m group,MφCD14、CD64、TLR4、CD11b、CD18、CD80、MHC-Ⅱexpression level of the 4200 m group were obviously decreased(P<0.05);After exposing to hypoxia for 30 days,in comparison with the 400 m group: The concentration of IL-6、TNF-α in the Mφ supernatant of the 4200 m and 2200 m group significantly increased(P<0.05).Conclusion After exposed to hypoxia at an altitude of 4200 m and 2200 m for 30 days,Mφ phagocytosis and killing ability both reduce;the function of cytokines secretion in Mφ changed and hypoxia prompted inflammatory reaction to a certain extent and changed the immunity ability of Mφ;the expression level of Mφ surface molecules were significantly changed.The phagocytosis and killing abilities of Mφwere suppressed a certain degree,prompting inflammatory reaction and changing the regulation of Mφ under the condition of low oxygen.
