Objective To investigate whether there is differences in the expression of follicle stimulating hormone receptor(FSHR)and stem cell factor(SCF)in mature and immature follicles,and to further explore whether the expression of FSHR and SCF in granulosa cells is related to follicular maturation.To study the effects of different concentrations of recombinant follicle stimulating hormone(rFSH)on SCF mRNA and protein expression in cultured human ovarian granulosa cells in vitro.To study a possible mechanism of FSH regulating SCF secretion in human ovarian granulosa cells.Methods Experimental study.The granulosa cells of patients undergoing intracytoplasmic sperm injection(ICSI)were collected and divided into two groups(mature follicles and immature follicles group).The expression of FSHR and SCF in granulosa cells,isolated from mature follicles and immature follicles,were detected.The primary GCs collected from follicular fluid(IVF)were divided into 6 groups and cultured with 10IU/L,25 IU/L,50 IU/L,100 IU/L,200 IU/L of rFSH(Gonal-F)and a control group(normal cultured),separately.The cultured GCs of intervention group and control group were subjected to treatments with cAMP or an inhibitor of PKA(H-89).GCs were cultured in control medium or in the presence of rFSH,H-89,cAMP,cAMP and H89 or rFSH and H89.The protein and mRNA expression of FSHR and SCF in GCs was detected by using Immunofluorescence,reverse transcriptionpolymerase chain reaction(RT-PCR),and Western blot.Results FSHR and SCF in GCs derived from immature follicles were lower secreted than those in GCs derived from mature follicles.With the rFSH treatments,SCF had a strong expression in primary cultured GCs.SCF was increased by cAMP and H-89 significantly inhibited the effects of both rFSH and cAMP on the expression of SCF mRNA and protein.Conclusions In human GCs,FSH may up-regulates the expression of SCF via the cAMP/PKA pathway to promote the follicle growth and development and the oocytes maturation.
