Objective To explore the effect of Cyclophilin A(Cyp A)on the growth and metastasis of laryngeal squamous cell carcinoma in nude mice,through the technology of RNA interference.Methods We constructed lentiviral vector-mediated shCyp A to down-regulate Cyp A,and infected hep2 cells.Fifteen mice were completely randomly divided into 3 groups(5 mice in each group): control group,NC ShRNA group,and Cyp A ShRNA group.The growth and lymph node metastasis of tumor were observed.The expression of Cyp A,CD147 and MMP-9 in tumor tissue were detected by immunohistochemistry.The expression of Cyp A,CD147 and MMP-9 mRNA and protein were detected by RT-PCR assay and Western blot.Results The gross tumor volume of Cyp A ShRNA group was significantly smaller than that of control group and NC shRNA group(P<0.05).Cyp A was found expressed in cytoplasm,whereas CD147 expressed in cytomembrane and MMP9 expressed in tumor stroma.The expressions of Cyp A,CD147 and MMP9 mRNA and protein were significantly decreased in CypA ShRNA group as compared to control group and NC shRNA group(P<0.05).Conclusion Down-regulation of Cyclophilin A can inhibit the growth and metastasis of laryngeal squamous cell carcinoma by regulating the expression of CD147 and MMP9.
