PART?BIOINFORMATICS ANALYSIS OFDIFFERENTIALLY EXPRESSED GENES IN HCCObjective To screen of the potential molecular markers for early detection and molecular targets for immunotherapy,differentially expressed genes were explored in HCC using bioinformatics methods.Methods Gene Expression Omnibus(GEO)microarray data related to liver cancer were download to analyze the correlation of those datasets,using JMP software,subsequently,differentially expressed genes(DEGs)were identified using Limma program package of language R for those GEO datasets.After that,GO enrichment and KEGG pathway analysis DEGs were performed,as well as protein interaction network(PPI)analysis.Moreover,the specify DEGs related to HCC were further screened out,combined with other cancer RNAseq data obtained from TCGA.Results RNAseq data verified that GPC3 was only up-regulated in liver cancer,while MBL2,SDS,SLCO1B3,TDO2,SAA4,SPP2 was downregulated.only the expression of SPP2 is related to lifetime of patients.Conclusion GPC3 might act as a target for immunotherapy,SPP2 should be the target for prognosis.PART?PRELIMINARY EFFECT IDENTIFICATION OFCSMD2 IN HCCObjective To explore the preliminary effect and mechanism of CSMD2,the evolutionary process and function annotation was analyzed,their differentially expression was detected in HCC and normal tissues.Method The sequences of CSMD2 in different animal species were searched from NCBI database,and phylogenetic tree was constructed by Tree Fam,then Function annotation of CSMD2 is analyzed by bioinformatics softwares,the expression of CSMD2 were detected in HCC? adjacent and normal tissues by real-time fluorescence quantitative PCR and IHC,then the correlation between expression and the clinicopathologic features.Results The expression of CSMD2 is higher in adjacent tissues than HCC tissues.The positive expression of CSMD2 in adjacent HCC tissues is 52.0%,while it is 84.0% in HCC tissues,the expression of CSMD2 in HCC tissues was not related to the clinical parameters like sex,age,preoperative AFP levels,BCLC staging,HBs Ag expression,the size and number of tumors.However,only lymph node metastasis is of significance(p<0.05).Conclusion The lack of CSMD2 expression in HCC and the tumor metastasis relevant.
