ObjectTo investigate effects of antidepressants on proliferation and differentiation of neural stem cells derived from human olfactory mucosa,for adding to growing evidences that neurogenesis may play a pivotal role in antidepressant therapy.MethodsTissue piece culture method and neurosphere method were applied to harvest olfactory cells and olfactory neural stem cells separately.Immunofluorescence was conducted to verify the specificity of neural stem cells and neural cells differentiated from them.CCK-8 and Edu were used to measure effects of antidepressant on proliferation of human olfactory neural stem cells.Western blot was applied to measure effects of antidepressant on differentiation into neural cells from neural stem cells.Results1.Immunofluoscence assay showed Nestin,TUJ1,positive in the treated cells.We successfully established culturing method of human olfactory neural stem cells.2.CCK-8 showed that fluoxetine could increase viability of olfactory neural stem cells at certain concentration.Edu assay exhibited that fluoxetine could promote proliferation of neural stem cells.3.Compared to control group,fluoxetine significantly increased expression of TUJ1 which means it could promote differentiation into neural cells from neural stem cells.Conclusion1.Culturing method of human olfactory neural stem cells is established and they have the capability to differentiate into neural cells.2.Antidepressants could increase neurogenesis of neural stem cells which is likely to be one of the mechanisms in antidepressant treatment.
