Objective:To test and analyse the levels of CD19+CD24hiCD38hiBreg and IL-10 in peripheral blood of the different stages of systemic lupus erythematosus?SLE?;To analyse their correlation with Th17 and Treg cells.Methods:We selected 50 SLE patients were initially treated and according to the conditions of the group,and 18 healthy cases were selected as control group.And the activity of the patients in the SLE group was divided into the stable group,the low activity group,the moderate activity group and the severe activity group according to the SLEDAI scores.The expression of the subsets of CD19+CD24hiCD38hiBreg,Treg and Th17 cells were detected by flow cytometry?FCM?.Analyze the expression of the IL-10 in serum of all experimental subjects by enzyme-linked immunosorbent assay?ELISA?.All the data were analyzed with SPSS 17.0.Results:The level of IL-10 in serum of patients with SLE?408.03±211.25?pg/ul were significantly higher than that of the normal controls?104.29±30.26?pg/ul?p<0.05?;IL-10,in the serum of patients with SLE were statistically significant difference between the stable group?185.02±69.27?pg/ul,the low activity group?306.98±65.09?pg/ul,the moderate activity group?431.37±127.50?pg/ul and the severe group?625.61±201.27?pg/ul?p<0.05?.The count of IL-10 were positively correlated with SLEDAI scores?r=0.779,p<0.01?.The count of CD19+CD24hiCD38hiBreg cells in the peripheral blood of the patients with SLE?1.66±1.61?/ul were significantly decreased than that of the normal controls?6.96±3.58?/ul?p<0.05?;CD19+CD24hiCD38hiBreg cells,in the peripheral blood of the patients with SLE were statistically significant difference between the stable group?3.58±2.20?/ul,the low activity group?1.85±1.32?/ul,the moderate activity group?1.10±0.56?/ul and the severe group?0.66±0.46?/ul?p<0.05?.The level of CD19+CD24hiCD38hiBreg cells were negatively correlated with SLEDAI scores?r=-0.703,p<0.01?and IL-10?r=-0.593,p<0.0?.The count of Th17 cells in the peripheral blood of the patients with SLE?40.76±48.37?/ul were significantly increased than that of the normal controls?6.45±3.98?/ul?p<0.05?;The count of Treg cells in the peripheral blood of the patients with SLE?4.94±4.19?/ul were significantly decreased than that of the normal controls?28.21±15.04?/ul?p<0.05?;Th17 cells,in the peripheral blood of the patients with SLE were statistically significant difference between the stable group?10.49±6.52?/ul,the low activity group?20.91±16.44?/ul,the moderate activity group?42.61±31.50?/ul and the severe group?76.65±68.90?/ul?p<0.05?;Treg cells,in the peripheral blood of the patients with SLE were statistically significant difference between the stable group?10.08±5.23?/ul,the low activity group?5.74±1.83?/ul,the moderate activity group?3.70±2.67?/ul and the severe group?1.83±2.00?/ul?p<0.05?.The level of Th17 cells were positively correlated with SLEDAI scores?r=0.756,p<0.01?and IL-10?r=0.607,p<0.0?and negatively correlated with CD19+CD24hiCD38hiBreg cells?r=-0.410,p<0.01?.The level of Treg cells were negatively correlated with SLEDAI scores?r=-0.733,p<0.01?and IL-10?r=-0.477,p<0.0?and positively correlated with CD19+CD24hiCD38hiBreg cells?r=0.568,p<0.01?.Conclusion:The subsets of CD19+CD24hiCD38hiBreg cells and IL-10 were participate in the pathogenesis of SLE,which is highly possible to regulate the Th17 and Treg cells.
