Background and Objective:Gallbladder cancer is a kind of primary malignant tumor in gallbladder,which is difficult to diagnose in early stage.,surgical radical resection rate is low,the chemotherapy is relatively insensitive to the poor prognosis,easy to recur and metastasis after surgery.So the study of gallbladder cancer for the treatment of new means is particularly important.Myricetin is a kind of flavonoids extracted from the leaves,dermis and roots of the genus Camelliae,which can be used for various treatments such as anti-inflammatory,anti-oxidation,analgesic,lowering blood sugar and protecting the liver.In recent years,various studies have shown that the drug myricetin in the anti-cancer also has a significant effect.Therefore,the aim of this study is to investigate the effect of myriophyllum on the apoptosis of gallbladder carcinoma in vitro and its mechanism,and to provide a new approach for chemotherapy of gallbladder carcinoma.Methods:This study was done by in vitro cell experiment.The experimental group(adding myricetin)and control group(without adding myricetin)were compared.CCK8 assay and monoclonal experiment were used to observe the inhibitory effect of different concentrations of myricetin on the proliferation of NOZ cells.The changes of NOZ cells apoptosis in different concentration group and control group were analyzed by flow cytometry.Western blot was used to detect the expression of related proteins in mitochondrial apoptosis pathway before and after the action of myricetinResults:Myricetin can significantly inhibit the proliferation of NOZ cells.The results of CCK8 kit method showed that the inhibition of cell proliferation was more obvious with the increase of myricetin concentration and the prolongation of time.Myricetin can inhibit NOZ cells and induce cell apoptosis.The results of flow cytometry showed that the apoptosis rate increased obviously with the increase of myricetin concentration,and the late apoptosis rate was more significant.The cells showed significant morphological changes under the microscope.Myricetin promotes NOZ cell apoptosis by regulating the expression of Bax,Cytc,PARP and Bcl-2 protein.The results of Western blotting showed that the protein expression of Bax,Cytc and PARP increased with the increase of myricetin concentration in a dose-dependent manner,while the expression of Bcl-2 protein decreased.Conclusion:Myricetesin has a good inhibitory effect on NOZ cells in vitro and can induce cell apoptosis,and its effect is time and dose dependent.
