| Objective: To study the protective effects of sevoflurane postconditioning to myocardial ischemia-reperfusion injury and prove that the hemeoxygenase-1(HO-1)play an important role in the way.Methods: 30 healthy male SD rats weighted 230~260g,were labeled to number in random way and divided randomly into 5 groups(n=6): sham operation group(S group),ischemia reperfusion group(I/R group),the sevoflurane postconditioning group(SP group)and sevoflurane postconditioning with Zinc protoporphyrin group(SP+Znpp group),Znpp group(Znpp group).The S group thread under the left descending coronary artery(LAD)without ligation with silk thread,whereas the I/R group,SP group,SP+Znpp group and Znpp group ligatured the LAD with 30 minutes,then loosed the thread and took the reperfusion with 120 minutes.The I/R group,Zn group respectively injected 1ml normal saline(NS)and Znpp diluent with NS 1 ml(5ug/kg),and inhaled pure oxygen at 15 minutes before reperfusion.The SP and SP+Znpp group respectively injected saline NS 1ml and Znpp diluent 1ml(5ug/Kg),and inhaled 3% sevoflurane at 15 minutes before reperfusion.After 120 minutes’ reperfusion,(1)Drawing blood from artery and measuring CK-MB content in serum with enzyme linked immunosorbent assay(ELISA).(2)Taking myocardium ischemia tissue and measuring SOD activity and MDA content in the tissue.(3)By using Western Blot assay to measure HO-1 protein expression level in myocardium ischemic tissue.Results: Compared with S group,the CK-MB,MDA content and HO-1 protein expression level of other groups were significantly increased,SOD activity decreased obviously,with statistically significant difference(P < 0.05);Compared with I/R group,the CK-MB,MDA content of the SP group lowered obviously,HO-1 level,SOD activity increased significantly,with statistically significant difference(P < 0.05);Compared with SP group,the c Tn-I,MDA content of the SP+Zn PP group were significantly increased,HO-1 level,SOD activity lowered obviously,with statistically significant difference(P < 0.05).Conclusion:Sevoflurane postconditioning have protective effects to the myocardial ischemia-reperfusion injury and HO-1 plays a key role in this protective function. |
