Objective:Evaluate the effect of different dosages monascin on hematoma and brain edema volumes after intracerebral hemorrhage rats by MRI.At the same time to observe its effect on neurological evaluation,brain water content,blood brain barrier permeability and the hematoma volume.To observe the protective effect of monascin after ICH in rats.Methods:164 adult male Sprague-Dawley rats were randomly divided into sham,vehicle monascin groups with low dosages(1 mg/kg/day),middle dosages(5 mg/kg/day)and high dosages(10 mg/kg/day)respectively.Each group was equally divided into three subgroups at 1,3,7 days after surgery.The right basal ganglia of rats were injected with collagenase IV 0.4 U to build ICH models.All experimental animals were given different dosages monascin or the same amount of 0.9% sodium chloride solution twice a day.Six rats of each group were used to evaluate neurological function scores and MRI at each observation point following postoperative.T2* WI and SWI were used for estimating hematoma volumes,T2 WI and T2 Flair for edema volumes around the hematoma.At 7days rats were sacrificed and the brains were collected.The remaining rats per group were sacrificed for brain specimen collection at 1,3 d after ICH,to examine western blot,the brain water content,blood brain barrier permeability and the hematoma volume.Results:(1)Compared with vehicle group,all the expressions of PPAR? and Nrf2 protein showed a signficant increase after intervened by 10 mg/kg monascin at 1,and 3 days after ICH.(2)Compared with vehicle group,after 24 h,all ICH animals demonstrated significant neurological deficits.The high dosage monascin significantly improved neurological deficits,reduced the hemoglobin levels in 1-7 days after ICH.And the same to the middle dosage monascin animals at 3-7 days.(3)The high dosage monascin reduced brain edema at days 1-3 after surgery,the middle dosage monascin rats also reduced at 3 days,but the effect of high dosage intervention group was more obvious.(4)Compared with vehicle group,the blood brain barrier permeability obvious damage after ICH.But the Evans blue content significantly decreased at 1-3 days after the high dosage monascin.(5)The hematoma and the perihematomal edema were apparent in the first 24 h after ICH,peaked on 3 days,then declined.ICH + high dosage monascin rats can significantly deflated the hematoma volume at 1,3,7 days and the edema volume were significantly decreased at 1-3 days after ICH.Meanwhile,the volume of hematoma were reduced at 3-7 days and the edema volume were slighted decreased at 3 days,which intervened by middle dosage monascin.Conclusion:Our study demonstrated that the high dosage monascin played a significant neuroprotective roles after ICH through reducing BBB permeability,edema and hematoma volume.
