Objective:To investigate the effects of interleukin-22 on TGF-? 1 induced epithelial-to-mesenchymal transition in rat hepatocytes.Methods:The BRL-3A cells were randomly divided into 5 groups: normal control group,model control group and the different concentration of recombinant IL-22 treated groups(1ng/ml,10 ng/ml,50ng/ml).2ng/ml TGF-?1 was applied to stimulate BRL-3A cells in all groups except the normal control group.The expression of E-cadherin and vimentin mRNA was detected by quantitative real time PCR.The expression of E-cadherin and vimentin protein was measured by Western blot.Results:(1)Compared with normal control group,the expression of E-cadherin mRNA [(0.81 + 0.06)vs(0.98 + 0.08),t = 2.868,P<0.05] and protein [(0.58 + 0.03)vs(0.77 + 0.06),t = 4.890,P<0.05] decreased,while the expression of vimentin mRNA [(1.24 + 0.02)vs(0.99 + 0.11),t =-3.805,P<0.05] and protein [(0.96 + 0.06)vs(0.67 + 0.08),t =-4.946,P<0.05] in model control group increased.(2)Compared with model control group,the expression of E-cadherin mRNA [(1.03 + 0.07)(1.31 + 0.02),(1.61 + 0.09)] and protein [(0.70 + 0.02),(0.80 + 0.03),(0.88 + 0.03)] increased(all P<0.05),while the expression of vimentin mRNA [(0.97 + 0.10),(0.57 + 0.03),(0.30 + 0.77)] and protein [(0.86 + 0.03),(0.74 + 0.02),(0.61 + 0.04)] in recombinant IL-22 of different concentrations intervention groups decreased.And the differences in the IL-22 intervention groups were statistically significant(all P<0.05).Conclusion:TGF-? 1 can induce an epithelial-to-mesenchymal transition in rat hepatocytes,and IL-22 is able to reverse this transition in a dose-dependent way.
