ObjectTo analyze the long non-coding RNA(Lnc RNA)that associated with the metastasis / recurrence / invasion of Hepatocellular Carcinoma(HCC)in different time periods and verify differentially expressed Lnc RNA.In order to further study Lnc RNA associated with the metastasis / recurrence / invasion of HCC and the molecular mechanisms of invasion and metastasis of HCC to provide basic information.Methodswe employed the Lnc RNAs microarray technology to study the Lnc RNAs expression profiles at genome-wide in hepatocellular carcinoma(HCC)tissue samples with early recurrence(less than 1 year,with invasion and metastasis out of liver,number=6)and late recurrence(longer than 2 years,without invasion and metastasis out of liver,number=6),which had different recurrent/metastatic potentials,by using normal liver tissue(number=6)as control to screen the dysregulated Lnc RNAs which are potentially involved in the recurrence,invasion and metastasis process of HCC.Coding-non-coding Gene Co-expression Network and Gene ontology and Pathway analysis were used to analysis the dysregulated Lnc RNAs.Furthermore,Select 15 Lnc RNAs of differentially expressed in hepatocellular carcinoma cell lines MHCC97 H and MHCC97 L with q-PCR test and verify.T test method for statistical analysis.Results1,170 LncRNAs were identified to differentially expressed between the early and late recurrence samples.These differentially expressed Lnc RNAs most distribution in the chromosome 2.We have classified the lnc NRAs as sense,antisense,intronic,bidirectional and intergenic,and analyzed the genomic context of differentially expressed Lnc RNAs.We found that all of the five categories could be examined in the differentially expressed Lnc RNAs.Strikingly,we found that nearly one third of them were long intergenic non-coding RNAs(linc RNAs)in all comparison.Common expression analysis and path analysis found that the Lnc RNAs associated protein-coding genes were enriched in cellular process,physiological process and metabolism.Fifteen differentially expressed Lnc RNAs selected were validated by q PCR in cell lines MHCC97 H and MHCC97 L,which have exactly same genetic background but with different invasion potentials.Nine of the selected Lnc RNAs are consistent with the microarray data.ConclusionsPart of LncRNAs may be associated with the metastasis / recurrence / invasion of Hepatocellular Carcinoma,and mainly distributed in chromosome 1 and 2.
