Objective:To investigate the expression of Activating transcription factor 3(ATF3)in prepuce tissues,and its relation to sporadically isolated hypospadias.Methods: 1.Specimen collecting: The experimental group was the redundant foreskin tissue in urethroplasty of sporadically isolatedhypospadias patients(there were 35 cases,10 cases were subjected to mild,15 cases were moderate and 10 cases were sever),and the control group was the redundant foreskin tissue in patients with redundant prepuce.2.Using the reverse transcription polymerase chain reaction(RT-PCR)to determinate the expression of ATF3 mRNA in the experimental group and the control group.3.Using the immunohistochemistry technique to identify the expression of ATF3 protein in the experimental group and the control group.4.Statistical processing: Data from the experiments were carried out strict inspection,through repeating measurementdata deflecting three standard deviationswere deletedSPSS12.0 statistics software was applied to input data and set up database.Count data was expressed as mean and standard deviation(+ /-),and measuring material was compared by F/t test.Count data was represent with positive cases or positive rate(%),positive rate was compared by chi-square test.The ? value is 0.05 as a standard test,the probability oftwo sides.Results: The relative expression of ATF3 in the experimental group and the control group are 0.54 ± 0.06,0.44 ± 0.07 respectively.The expression of ATF3 in sporadically isolatedhypospadiassignificantly increased compared with normal prepuce.Two groups of results comparison difference has the obvious significance(t=5.89,P=0.00).The differently related expression ofATF3 m RNA in patients with different severity was not significant(F=0.13,P=0.88).The positive rate of ATF3 protein detected by immunohistochemistry are 85.7%(30/35),15.0%(3/20)respectively in the experimental group and the control group.Comparing two groups,the expression rate of ATF protein positive showed statistical difference.The positive rate of ATF3 protein detected by immunohistochemistry in sever patients showed that the positive rate of ATF3 protein increased by degrees in patients with different severity,however there was not significant difference.Conclusions:The expression of ATF3 in the prepuce of sporadically isolatedhypospadias patient increase dramatically compared with patient withabundant prepuce including phimosis,which suggest a close relation between ATF3 and sporadicallyisolated hypospadias.Thisresearch provide a experimental basis for further research on signal transduction of ATF3 in hypospadias and ATF molecular basis,and provide scientific basis for the pathogenesisofhypospadias.
