Objective:?COPD?model of CCR6-/-mice induced by cigarette smoke and to observe the expression of CD80,CD1 a in dendritic cells?DCs?and helper T cells 17?Th17?,T cells?Treg?changes in lung tissue of mice,to further explore the pathogenesis of COPD.Methods: 20 C57BL/6 background CCR6-/-mice and 20 wild type C57BL/6 mice were randomly divided into 4 groups.CCR6-/-mice were treated with air control group?n = 10?,CCR6-/-mice were smoked Group?n = 10?;wild-type air control group?n = 10?,wild-type smoked group?n = 10?.The COPD mice model was established by the method of smoking.The mice in the COPD model were established using the smoke method.Four groups of mice were sacrificed within 24 hours after the last smoking in the fourth week?28days?and the lung tissue samples were collected.Lung tissue was taken as paraffin section,immunohistochemical staining and HE staining.The changes of CD80,CD1 a,Th17 and Treg in lung tissue were analyzed by immunohistochemistry.Results:?1?Cigarette smoke can induce mouse COPD formation,and CCR6-/-mice emphysema is better than wild type mice;?2?The lung tissue before and after smoked the expression of CD80 in the wild type mice was higher than that of the CCR6-/-mice,and the difference was statistically significant.Compared with their respective air control group,reduce the expression of CD80 in lung tissue of mice in the smoking group.The changes and changes rate of CD80 in lung tissue of wild type mice were significantly higher than those of CCR6-/-mice before and after smoking,and the difference was statistically significant;?3?the lung tissue before and after smoked the expression of CD1 a in wild type mice was higher than that of CCR6-/-mice,the difference was statistically significant,and compared with their respective air control group increased CD1 a expression in lung tissue of mice in the smoking group.The changes and changes rate of CD1 a in lung tissue of wild type mice were significantly higher than those of CCR6-/-mice before and after smoking,and the difference was statistically significant;?4?the lung tissue before and after smoked the expression of Th17 in wild type mice was higher than that of CCR6-/-mice,the difference was statistically significant,and compared with their respective air control group increased Th17 expression in lung tissue of mice in the smoking group.The changes and changes rate of Th17 in lung tissue of wild type mice were significantly higher than those of CCR6-/-mice before and after smoking,and the difference was statistically significant;?5?The lung tissue before and after smoked the expression of Treg in wild type mice were lower than those of CCR6-/-mice,the difference was statistically significant,and compared with their respective air control group decreased the expression of Treg in lung tissue of mice in the smoking group,the difference was statistically significant.The difference of Treg expression in lung tissue of wild type mice before and after smoking was less than that in CCR6-/-mice,and the difference was statistically significant,but after smoking,the rate of Treg change in wild mice was higher than that in CCR6-/-mice;?6?The ratio of Th17 / Treg in the lung tissue of the smoked group was higher than that of the control group,Indicating that there is Th17 / Treg imbalance in cigarette model induced by cigarette,and the mice in the wild type smoked group were higher than those in the CCR6-/-smoked Group,the difference was statistically significant.Conclusion: After CCR6 knockout,pulmonary inflammation and emphysema improved,indicating that CCR6 plays an important role in the pathogenesis of COPD,which may be related to the decrease in CCR6+ cell recruitment,especially DCs and Th17;Cigarette smoke results in an increase in i DCs and a decrease in m DCs,suggesting that DCs may play a vital role in the pathogenesis of COPD;DCs are presented by antigen,or the secretion of cytokines affects the T cell-mediated immune response,which may cause Th17 / Treg imbalance,Th17 / Treg imbalance in the occurrence and development of COPD may play an important role.
