Objective: To determine the role Smoothened,the downstream component of Shh signal,played in the Shh pathway of cultured mouse embryonic palatal mesenchymal cell and the effect of serum starvation on ciliated rate of the cell.Methods: Mouse embryonic palatal mesenchymal cell were extracted from mouse embryos with embryonic days of 13.5 to 15.5 and incubated with Shh,SAG and serum starvation medium.Smoothened was labeled with immunofluorescence to observe the effect of serum starvation on ciliated rate of the cell.Cultured cell were divided into control group,treatment group 1 and 2 which received Shh or SAG treatment respectively,while control group was given serum starvation medium only.Acetylated ?-tubulin and Smoothened was labeled to observe the changes in Smoothened accumulation.Results: The ciliated rate of cultured mouse embryonic palatal mesenchymal cell increased when underwent serum starvation.Smoothened was detected in the cilia of palatal mesenchymal cell in vitro when incubated with Shh and SAG while the cilia of the control group cell did not show Smoothened accumulation.Conclusion: Serum starvation can increase the ciliated rate of cultured mesenchymal cell.Mouse palatal mesenchymal cell extracted from mouse embryos with embryonic days of 13.5 to 15.5 is responsive to Shh signal and SAG treatment.
