Objective To observe the effect of scalp acupuncture on the expression of NF-kappa Bp65 mRNA and I kappa BmRNA in ischemic penumbra of acute MCAO rats,and explore the mechanism of scalp acupuncture in regulating the inflammatory response of acute ischemic cerebrovascular disease.Methods 100 male SD rats,20 rats were randomly selected as normal group,the rats with middle cerebral artery occlusion of MCA occlusion(middle cerebral artery occlusio,MCAO)model in MCAO rats,confirming the successful preparation of the model will accord with standard rat model by neural function were randomly divided into model group(N=20),scalp acupuncture group(N=20)and pyrrolidine dithiocarbamate(pyrrolidinedithio two carbamate,PDTC)group(N=20).The normal group of SD rats,feeding,without any intervention,only to do the same with the intervention group after modeling the crawl and fixed;no treatment group,only to do the same with the intervention group crawling and fixed;scalp acupuncture group in modeling after 6h began to take intervention.Double D to 15~20 DEG relay rapid penetration,each needle each with 100 RPM twist 1min,1 /10 min,a needle for 30 minutes,1 times /d,7d;in the PDTC group after the modeling 6h began to intraperitoneal injection intervention,intraperitoneal injection of PDTC 100 mg.kg-1/d,7d.The intervention of rats after 7d,neurological deficit score,were sacrificed from the brain,rapid separation of the left hemisphere,cut the ischemic penumbra,using real-time reverse transcriptase polymerase chain reaction(Real time RT-PCR)was used to detect the expression of NF-kappa Bp65 mRNA,I kappa BmRNA;pathological change of ischemia rats penumbra pathology.Result(1)the effect of scalp penetration needling on the neurological impairment score of MCAO ratsAfter modeling,the rats were awake at about 1H,the normal group had no neurological deficits before intervention;compared with the normal group,model group,scalp acupuncture group and PDTC group showed the symptoms of neurological deficit(P<0.05),and the 3 group of neural function defect score was no significant difference(P>0.05);intervention compared with the model group,scalp acupuncture group and the scores of rats in PDTC group were significantly decreased neurological deficit(P<0.05);compared with PDTC group,scalp acupuncture group of neurological score was significantly lower(P<0.05).(2)pathological analysis of ischemic penumbra in rats of each groupNormal group: normal cell morphology,neat,no inflammatory cell infiltration;model group: the nerve cells are scarce,loosely arranged,nerve cell shrinkage,degeneration;PDTC group: compared with the model group,the infiltration of inflammatory cells reduced interstitial edema;scalp acupuncture group: compared with PDTC group,the infiltration of inflammatory cells further reduced,further reduce the interstitial edema.(3)the effect of scalp acupuncture on NF-kappa Bp65 mRNA of neurons in ischemic penumbra of MCAO ratsCompared with the normal group,model group,expression of NF-kappa Bp65 mRNA increased significantly(P<0.05);compared with the model group,scalp acupuncture group and PDTC group the expression of NF-K Bp65 mRNA were significantly decreased(P<0.05);compared with PDTC group,scalp acupuncture group NF-kappa Bp65 mRNA expression had no significant difference(P> 0.05).(4)the effect of scalp acupuncture on I kappa BmRNA of neurons in ischemic penumbra of MCAO ratsCompared with the normal group,model group,I expression of kappa BmRNA decreased significantly(P<0.05);compared with the model group,scalp acupuncture group and PDTC group the expression of I K BmRNA were significantly increased(P<0.05);compared with PDTC group,scalp acupuncture group I kappa BmRNA expression had no statistical difference(P>0.05).Conclusion Expression of rat ischemic penumbra neurons of I kappa BmRNA,and inhibited NF-expression of NF kappa Bp65 mRNA,improve the ischemic penumbra neuronal injury is one of the mechanisms of scalp acupuncture regulation of inflammation in acute ischemic cerebrovascular disease reaction.
