The Expression And Clinical Significance Of Circ CD44-360 And CircCD44-594 In Bladder Cancer

Object: In this study,we surveyed the circCD44-360?hsa_circ₀021727?and circCD44-594?hsa_circ₀021728?expression profiles of bladder cancer and para-cancer tissues,and theirs correlation and effects on CD44 gene expression,aimed to new strategies for bladder cancer diagnosis and targeted therapy.Methods: We collected bladder cancer and para-cancer tissues specimens.After processing the specimens,we extracted the total RNA of bladder cancer and para-cancer tissues,then we obtained the target RNA by reverse transcription.To confirm the differential expression of circCD44-360 and circCD44-594,the target RNA were examined by qRT-PCR.To find out the relationship between the regulating mechanism we analyzed circCD44-360 and circCD44-594 respectively with parent gene CD44 messenger RNA?mRNA?.Results:1 Up-regulation of both circCD44-360 and circCD44-594 expression in BC.qPT-PCR analysis results show that both circCD44-360 and circCD44-594 expression was significantly upregulated in BC,compared to para-cancer tissues.Taken together,our results demonstrated the upregulation of circ CD44-360 and circCD44-594 in BC.2 Up-regulation of CD44 mRNA expression in BC.qPT-PCR analysis results show that CD44 mRNA expression was significantly upregulated in BC,compared to para-cancer tissues.Taken together,our results demonstrated the upregulation of CD44 mRNA in BC.3 Both circCD44-360 and circCD44-594 with parent gene CD44 is linear positive correlation.Using qPT-PCR analysis results for statistical analysis,we confirmed that circCD44-360 and its parent gene CD44 Pearson's correlation was linear positive correlation,R=0.5702,P=0.0087.Also circCD44-594 and its parent gene CD44 Pearson's correlation was linear positive correlation,R=0.4474,P=0.0420.Conclusions:1 CircCD44-360 expression is significantly upregulated in BC.2 CircCD44-594 expression is significantly upregulated in BC.3 CircCD44-360,circCD44-594 and the expression of CD44 is positive correlation in BC,so they may promote BC development and progression by regulating the expression of CD44 gene.
