Acute respiratory infections(ARI)is currently the main cause of death of acute infectious diseases in China and around the world.It has become a global public health problem.ARI can be caused by a variety of pathogenic microorganisms,in which the virus is the main cause of ARI pathogens.Therefore,carrying out ARI pathogens detection and separation has important practical significance for ARI epidemic monitoring and prevention.In this study,the pharyngeal swab samples from ARI patients in Panjin area from April 2013 to March 2014 were used as experimental materials to analyze the virus pathogens,and the epidemiological signs of new infectious diseases,major infectious diseases and early cases were analyzed and prejudged to avoid sudden public health events.The results are as follows:1.By using nucleic acid extraction and quantitative real-time PCR methods,856 throat swabs samples from ARI patients were detected for influenza virus(H1N1,H3N2,IVB),syncytial virus(RSV),parainfluenza virus(PIV1,PIV2,PIV3),adenovirus(AdV),partial pulmonary virus(MPV)and Rhinovirus(HRV).The results indicated that:(1)Influenza virus,parainfluenza virus,rhinovirus and adenovirus were the main pathogens causing ARI in this region.The positive rates of them were 72.38%,5.71%,6.67% and 10.48% respectively.(2)67 strains of H1N1 and 9 strains of H3N2 were detected from positive samples of influenza virus,accounting for 88.16% and 11.84% of the positive samples respectively,but no influenza B virus was detected.2.In 2013 and 2014,the prevalence of influenza virus in Panjin area was the following: winter was the highest incidence and lowest in summer.The age of onset was mainly in the age group of 5-14 years old,and the lowest in the 25-59 age group.Mainly sporadic cases,no epidemic outbreak of influenza,and the new H1N1 was the predominant strain,while H3N2 subtype less appeared.3.Through comparing the MDCK cell isolation and chicken embryo isolation of the influenza virus in Panjin area,it was found that the viruses were still less sensitive to chicken embryo than to MDCK cells.However,MDCK cells cannot be used directly for virus isolation in influenza virus vaccine production.So,MDCK and chicken embryo isolation need to be used in combination and the isolated strains be submitted to national influenza center for vaccine production.In summary,by using molecular biology method,virus pathogens were detected and analyzed for samples from AIR patients in Panjin area from April 2013 to March 2014,and the main viruses in samples and their occurrence regularity were preliminarily understood.Also,an influenza virus isolation method in combination of MDCK and chicken embryo was developed.The research results provided some scientific basis for clinic drug treatment,vaccine preparation,precaution of sudden public health events,decision and on-site treatment.
