| Objectives:To investigate the effects of hypoxia on the secretion of vascular factorsand migration ability of lung cancer A549 cells.Methods:(1)The lung cancer A549 cells were divided into the experimental group and the control group.The experimental group was cultured in hypoxic environment,and the control group was cultured in the normoxic environment.(2)2×105 cells were inoculated in the 12 plates of the experimental group and the control group,and the supernatant of the cells was collected after culturing 72 h.The expression levels of HIF-1,VEGF,TNF-a,ANG-2,TGF-alpha and TGF-beta in supernatant were detected by ELISA.Results:(1)Compared with the control group,the levels of HIF-1α,VEGF,TNF-a and ANG-2 in the supernatant of the experimental group were higher than those in the control group(P < 0.05),and the levels of TGF-alpha and TGF-beta in the experimental group were lower than those in the control group(P < 0.05).HIF-1α,and other signaling pathways can regulate certain vascular factors in a variety of ways,Ang-2 experimental group compared with the control group,the difference was not statistically significant.(2)Compared with the control group,the migration rate of 0h,6h and 20 h in the experimental group was accelerated,and the coverage time of the scratch was shortened.Conclusion:(1)Hypoxia can regulate the expression levels of many vascular factors secreted by A549 cells and participate in the mechanism of lung cancer.(2)Lung cancer A549 cells have stronger migration ability in hypoxic microenvironment than normal oxygen,and hypoxia can promote the proliferation of lung cancer cells. |
