Objective:To primarily explore the contributing mechanisms of EVI gene in children acute lymphoblastic leukemia.Method:1.30 children with new diagnosed ALL,aged 16 or less,were consecutively enrolled from May 1,2015 to September 30,2016 in Department of Pediatrics Hematology of the Affiliated Hospital of Qingdao University.Their Bone marrow was collected and compared with donated bone marrow of 6 healthy Children as normal control.Total RNA was extracted from bone marrow samples.The relative expression level of EVI1 m RNA was detected by quantitative qRT-PCR.The relative abundance of EVI1 mRNA expression was compared between the ALL group and the normal control group.2.Two age and disease severity matched children were selected,and bone marrow transcripttome gene sequencing was done between the EVI1 gene(+)and EVI1 gene(-)ALL children.We preliminary studied the role of EVI1 gene in children ALL pathogenesis through comparing the difference in gene expression.Result:1.The expression of EVI1 gene in bone marrow cells of ALL children and normal children were both tested.Level of EVI1 mRNA was 17.7 to 20.1,median 18.9,in normal children in contrast with 0.75 to 4096,median 934.65,which was significantly different(P= 0.01).Age,sex and disease severity matched samples with appropriate RNA concentration were selected for further transcriptome gene sequencing test.2.A 638 bp transcribed version of EVI1 gene was detected in EVI1(+)ALL children,which was not found in EVI1(-)ALL children.3.Comparing the difference in gene expression number,a total of 1387 genes were up-regulated and 1097 genes were down-regulated in EVI1-positive ALL children compared with EVI1-negative ALL children.Further analysis of the up-regulated genes showed that,(1)GATA2,BCL-xL,C / EBP?,which were related to hematopoietic stem cells and are regulated by EVI1 gene were included;(2)Myeloid lineage associated genessuch as CD34,CD13,CD13,CD116,CD114,CD124,CD125,CD126 and CD11 b were significantly up-regulated in EVI1(+)ALL children;(3)and so were the ECM receptor expression related genes.Conclusion:1.The expression of EVI1 gene in bone marrow cells of ALL children and normal children were both tested.Level of EVI1 mRNA was 17.7 to 20.1,median 18.9,in normal children in contrast with 0.75 to 4096,median 934.65,which was significantly different(P= 0.01).Age,sex and disease severity matched samples with appropriate RNA concentration were selected for further transcriptome gene sequencing test.2.A 638 bp transcribed version of EVI1 gene was detected in EVI1(+)ALL children,which was not found in EVI1(-)ALL children.3.Comparing the difference in gene expression number,a total of 1387 genes were up-regulated and 1097 genes were down-regulated in EVI1-positive ALL children compared with EVI1-negative ALL children.Further analysis of the up-regulated genes showed that,(1)GATA2,BCL-xL,C / EBP?,which were related to hematopoietic stem cells and are regulated by EVI1 gene were included;(2)Myeloid lineage associated genes such as CD34,CD13,CD13,CD116,CD114,CD124,CD125,CD126 and CD11 b were significantly up-regulated in EVI1(+)ALL children;(3)and so were the ECM receptor expression related genes.
