| [Objective] We explored the anti-tumor mechanism of fangchinoline(FAN)in triple negative breast cancer(TNBC).In order to achieve this goal,we discussed that the FAN could promote the apoptosis of TNBC cell(MDA-MB-231).Then we studied how FAN can inhibited the expression of protein which include PI3 K,AKT,mTOR protein and the phosphorylation of PI3 K,and AKT,mTOR protein.The last we studied wether FAN could inhibit the growth nude mices’ tumor.[Methods] First we cultivated MDA-MB-231 cells in cell incubator,and detected the inhibition of FAN in MDA-MB-231 cells by Alamarblue.Then we caculated the IC50 of the concentrate of FAN.We tested cell migration by the wound scratch assay and detected cell apoptosis by the Flow cytometry.Then we detected the expression of PI3 K,AKT,mTOR protein and the phosphorylation of PI3 K,AKT,mTOR protein by western blot.Finally we explored whether FAN could inhibit the growth nude mices’ tumor.Which were transplanted the breast cancer.[Results] FAN could inhibit the activity of breast cancer cells(MDA-MB-231),and the IC50 concertrate is 6.25μmol/L.The migration of breast cancer cells(MDA-MB-231)were inhibited by FAN,with the increased of the concentration of FAN,the inhibition was more obvious.FAN could induce the MDA-MB-231 cells apoptosis,with the increased of the concentration of FAN,the apoptosis rate of cell increased,and the apoptosis rate were positively correlated with the concentrate of FAN.The experimental results of Western Blot show that FAN could reduce expression of PI3 K,AKT,mTOR protein and the phosphorylation of PI3 K,AKT,mTOR protein,and the result was influenced by the the concentrate of FAN.The expression of ralated proteins was declined with the concentrate of FAN reduced.At last we could see that FAN could inhibit the growth nude mices’ tumor.which were transplanted the breast cancer.[Conclusions] FAN could reduce TNBC cells(MDA-MB-231)activity,accelerate them apoptosis,inhibit their migration and degrate the expression of PI3 K,AKT,mTOR protein and the phosphorylation of PI3 K,AKT,mTOR protein.FAN could inhibit the PI3K/AKT/mTOR signaling pathway,which inhibited the proliferation and migration of TNBC cells,inducing apoptosis,FAN could slow down the growth of mices’ tumor.It may have anti-tumor effect. |
