Objective: Sleep disordered breathing(SDB)has been closely associated with systemic diseases in recent years.Obstructive sleep apnea hypopnea syndrome(OSAHS)is characterized by frequent dyspnea or apnea in the sleep state,and the recurrent disorder in the body hypoxia causes systemic damage to multiple systems,multiple organs.According to the pathophysiological characteristics of OSAHS,a chronic intermittent hypoxia animal model with its characteristics was established to provide a reliable experimental platform for the study of sleep and respiratory diseases.Through the chronic intermittent hypoxia modle,to investigate the effects of adipokines that was induced by chronic intermittent hypoxia and reoxygenation on glucose metabolism and the expression of glucose transporter 2(GLUT-2),insulin receptor substrate 2(IRS-2)in renal.Methods: Twenty-four male S-D rats were randomly divided into control group,chronic intermittent hypoxia group(CIH),chronic intermittent hypoxia reoxygenation group(RH).CIH group and RH group were modeled in the chronic intermittent hypoxia animal model for 8 hours every day.After 4 weeks,rats in the RH group were conventional feed for 3 weeks.At the end of the model to extract the arterial blood immediately for the blood gas analysis.Fasting after 12 h intravenous blood centrifugation to take supernatant for using.Blood glucose and serum insulin were respectively detected by peripheral and radioimmunoassay,The serum levels of serum leptin and free fatty acid(FFA)were measured by enzyme-linked immunosorbent assay(ELISA).Remove the rat kidney tissue,the m RNA expression of GLUT-2 and IRS-2 in rat kidney were detected by q PCR method.Protein expression of GLUT-2 and IRS-2 were detected by Western blotting(WB)and immunohistochemistry(IHC).Results:1.The oxygen saturation of rats in control group was ? 95%,the oxygen saturation of CIH group was less than or equal to 85%,and the oxygen saturation degree of RH group was more than 86%;2.The levels of fasting blood glucose,serum insulin and insulin resistance in CIH group were significantly higher than those in control group and RH group(P <0.05).The RH group was higher than control group(P <0.05);3.The concentration of Leptin and FFA in CIH was higher than that in control group and RH group(P <0.05);RH group was higher than control group(P <0.05);4.The m RNA expression of GLUT-2 and IRS-2 in CIH group were higher than that in control group and RH group(P <0.05);The RH group was higher than control group(P <0.05);5.The protein expression of GLUT-2 and IRS-2 in CIH group was higher than that in control group and RH group(P <0.05);The RH group was higher than control group(P <0.05);Conclusion: Effects of chronic intermittent hypoxia on fasting blood glucose,insulin,leptin and FFA in rats increased the expression of GLUT-2 and IRS-2 in rat kidney,enhanced insulin resistance and reduced insulin sensitivity.
