OBJECTIVE: To investigate the effect of resveratrol(RES)pretreatment on cognitive function after sevoflurane anesthesia in neonatal SD rats.Methods: Newborn SD rats were randomly divided into control group(Con),resveratrol(RES)group,sevoflurane group(Sevo group),sevoflurane + resveratrol(Sevo + RES)group.SD rats were exposed to 2.5% sevoflurane 2 h daily for 3consecutive days in a self-made anesthesia box to establish a sevoflurane exposure model,and were treated with resveratrol at the dose of 30mg/kg or vehicle 30 min before the sevoflurane anesthesia from postnatal days 6(P6)to P8.The control group and the resveratrol group were exposed to the same conditions as the other conditions except sevoflurane.Rats were killed on P8,and the brain tissue were harvested.The morphological changes of brain tissue were observed by HE staining,Real-time polymerase chain reaction(RT-PCR)was used to detect the expression of Sirt1,PGC-1?and FOXO3? mRNA in hippocampus,The expression of Sirt1 and Caspase3 protein in hippocampus was detected by western blotting,and the changes of SOD and MDA activity in Hippocampal Homogenate by Chemical Method of Superoxide Dismutase(SOD)and Malonaldehyde(MDA)Kits.In another separate experiment,cognitive functions were evaluated by Morris water maze on P30.Results: Compared with group Con,the nerve tissue of Sevo group was obviously loose,edema,partial cytoplasmic dissolution,nuclear condensation,fragmentation,andSevo + RES group also showed cell edema,nuclear pyknosis and other pathological morphology,but significantly reduced than the Sevo group;the expression of Sirt1,PGC-1? and FOXO3? mRNA and SOD activity weredecreased(P <0.05),MDA level was increased(P <0.05)in Sevo group compared with group C;but compared with Sevo group,the expression of Sirt1,PGC-1? and FOXO3? m RNA and SOD activity in Sevo + RES group were significantly increased(P <0.05)and MDA level was decreased(P <0.05).Western blot analysis showed that Sirt1 expression in Sevo group and Sevo +RES group was lower than that in group Con(P <0.05),and Caspase3 in group Sevo was significantly increased compared with group Con(P <0.05),but compared with Sevo group,the expression of Sirt1 increased(P <0.05)and Caspase3 expression decreased(P <0.05)in Sevo+RES group.Conclusion: Resveratrol can reduce the level of oxidative stress and reduce the brain injury of neonatal SD rats induced by sevoflurane exposure.However,regardless of whether pretreatment with resveratrol,sevoflurane Long-term learning and memory ability without significant damage.
