Objective:1.To verify the effect of ZAG on the expression of insulin resistance-related adipokines,and try to clarify the role of ZAG in the development of insulin resistance.2.To verify the regulatory effect of ZAG on Akt-mediated insulin signaling pathway from cell level.Methods:The levels of IL-6,IL-6,IL-6 and IL-6 were detected by ELISA in the supernatant,cell m RNA and cellular protein of the 3L3-T1 adipocyte group,the pre-3L3-T1 adipocyte insulin resistance model and the ZAG-TNF-?,the expression of IL-6,TNF-?,adiponectin,leptin,PP2 A and Akt mRNA were detected by RT-PCR.The expression of adiponectin,leptin and PP2 A were detected by Western-blotting,P-Akt protein expression,and statistical,obtained correlation.Results:The expression of IL-6,TNF-?,adiponectin,leptin,PP2 A and Akt in insulin resistance group was higher than that in control group and ZAG group,and the expression of adiponectin and Akt was lower than that of control group and ZAG group.ZAG improved insulin resistance The expression of IL-6,TNF-?,leptin and PP2 A was lower than that of insulin resistance group,and the expression of Akt and adiponectin was higher than that of insulin control group,but still lower than that of normal control group.Conclusion:1.ZAG can improve insulin resistance,its play a way is multifaceted,not a single;2.ZAG can improve the expression of insulin resistance in insulin resistance;3.ZAG can directly enhance the insulin sensitivity from the insulin signaling pathway;4.ZAG on the insulin signaling pathway of its negative regulatory role of PP2 A can also play a role in down-regulation.
